dc.description.abstractBei der Aspartylglukosaminurie handelt es sich um eine lysosomale Speicherkrankheit. Die Mutationen AGU-Fin-major und T122K führen zu einer reduzierten Prozessierung und Aktivität der Aspartylglukosaminidase. Durch die eingeschränkte Aktivität dieses Enzyms kommt es zur Akkumulation von Aspartylglukosamin in den Lysosomen. Die Patienten werden symptomfrei geboren und im Verlauf ihres Lebens treten dann progressiv geistige und physische Einschränkungen auf. Das betroffene Enzym, die Aspartylglukosaminidase wird standardmäßig als enzymatisch inaktives Vorläuferpeptid synthetisiert und im endoplasmatischen Retikulum autokatalytisch gespalten. Das aktive Enzym bildet ein Tetramer, welches anschließend im Lysosom getrimmt wird. Die in dieser Arbeit untersuchte Mutation AGU-Fin-major entsteht durch einen Austausch von Cystein163 gegen Serin und Arginin161 gegen Glutamin. Die Mutation T122K wird durch den Austausch von Threonin122 gegen Lysin ausgelöst. Diese beiden Mutationen erzeugen AGA-Vorläuferenzyme, jedoch findet kaum Prozessierung in deren Untereinheiten statt.Diese Arbeit hatte das Ziel herauszufinden, ob bestimmte Substanzen Einfluss auf die Prozessierung des Vorläuferenzyms oder die Aktivität von AGA in Patientenfibroblasten haben. Zur Anwendung kamen rekombinantes Hsp70, Celastrol, Arimoclomol und MG 115, welche die Hitzeschock-Antwort auf unterschiedliche Weise aktivieren. Es sollte untersucht werden, inwieweit das AGA-Vorläuferenzym durch die Behandlung vermehrt in seine Untereinheiten prozessiert wird und somit die Bildung des aktiven Enzyms gefördert wurde. Eine erhöhte Prozessierung konnte in keinem der Versuche nachgewiesen werden. Die AGA-Aktivität dagegen konnte durch Celastrol und MG 115 bei den AGU-Fin-major Patientenfibroblasten signifikant gesteigert werden. Celastrol hat bei den AGU-Fin-major Patientenfibroblasten zudem zu einer Veränderung der Lysosomenmorphologie geführt. Unter Arimoclomol wurde die AGA-Aktivität von Patientenfibroblasten mit T122K Mutation überraschenderweise signifikant gesenkt. Die Behandlung von T122K Patientenzellen mit Hsp70 führte dagegen zu einer 50%igen AGA-Aktivitätssteigerung. Der Wirkungsgrad der hier an AGU-Patientenfibroblasten getesteten Substanzen, weist darauf hin, dass sie sich als mögliche Therapeutika zur Behandlung von Patienten mit Aspartylglukosaminurie als nicht ausreichend effektiv und sehr abhängig von der Art der Mutation erweisen.de_DE
dc.description.abstractAspartylglucosaminuria (AGU) is a lysosomal storage disease. The mutations AGU-Fin-major and T122K result in a reduced processing and activity of aspartylglucosaminidase. Limited activity of this enzyme leads to accumulation of aspartylglucosamine and other glycoasparagines in lysosomes. Patients are born symptom-free, but progressive mental retardation and physical decline occur throughout their lives. The affected enzyme aspartylglucosaminidase is synthesized as an enzymatically inactive precursor peptide which is further cleaved autocatalytically in the endoplasmic reticulum. The active enzyme forms a tetramer, which is later on trimmed in the lysosome.The first mutation studied in this work, AGU-Fin-major, results from an exchange of cysteine163 to serine and arginine161 to glutamine. The second mutation, T122K, is an exchange of threonine122 to lysine. Each of these mutations generate AGA precursor enzymes whose processing in the subunits barely takes place.The aim of this study was to characterize the effects of certain substances on the processing of the AGA precursor or on the enzyme activity of AGA in AGU patient fibroblasts. Hsp70, Celastrol, Arimoclomol and MG 115, which activate the heat shock response in different ways, were used. The focus was laid on to what extent the AGA precursor enzyme is converted to its subunits by the treatment and if the formation of the active enzyme is promoted. An enhanced processing was not detected in any of the experiments. However, AGA activity was significantly increased by treatment with Celastrol and MG115 in AGU-Fin-major patient fibroblasts. Treatment with Celastrol also has changed the lysosomal morphology in these fibroblasts. When treated with Arimoclomol, AGA activity of patient fibroblasts with T122K mutation was significantly reduced. Treatment of T122K patient cells with Hsp70 resulted in a 50% increase of AGA activity. The substances tested in AGU patient fibroblasts seem not to be effective enough to be used as potential therapeutics for treatment of patients with AGU. However, the effect of the tested substances clearly depend on which kind of AGU mutation is present.en
