dc.description.abstractDer intestinale Kaliumtransport wird durch verschiedene Second Messenger Systeme (v.a. cAMP und Ca2+) reguliert. Ein andererSignalweg, der Einfluss auf z.B. den Na+- und Cl--Transport nimmt, ist das Zellvolumen. Volumensensitive K+-Kanäle kommen in derbasolateralen Membran von Kolonkryptenzellen vor. Deshalb wurde untersucht, welchen Einfluss Veränderungen im Zellvolumen auf dentransepithelialen K+-Transport haben. Eine Zellschwellung wurde durch ein hypoosmolares Medium induziert. Veränderungen im Ionentransport am distalen und proximalen Kolonder Ratte wurden mit Hilfe von unidirektionalen Flüssen, Efflux- und Uptake-Versuchen erfasst; als Marker für K+ wurde 86Rb+ eingesetzt. Am distalen Kolon induzierte eine Zellschwellung, ausgelöst durch ein hypoosmolares Medium, eine Netto-K+-Sekretion. Im Unterschieddazu führte die Inkubation des Gewebes in einem hyperosmolaren Medium - vermittelt über eine Zellschrumpfung - zu einer Hemmung desresorptiven Flusses (Jms) von K+, insbesondere am distalen Kolon. Die Regulation des K+-Transportes durch das Zellvolumen ist nicht auf eine Änderung in den Aktivitäten von K+-Aufnahmemechanismenzurückzuführen, da weder ein hypo- noch ein hyperosmolares Medium einen Effekt auf den K+-Uptake zeigte. Stattdessen wurde derK+-Ausstrom verändert. Zellschwellung stimulierte den K+-Efflux in beiden Dickdarmabschnitten, während Zellschrumpfung ihn verminderte.Der apikale schwellungsinduzierte Efflux erwies sich als sensitiv gegenüber dem K+-Kanalblocker Ba2+ und der basolaterale gegenüberdem K+-Kanalblocker Quinin am distalen Kolon. Inkubation in einer Ca2+-freien Lösung mit 5 mmol.l-1 Mg2+ und Zugabe von La3+, einem unspezifischen Kationen-Kanalblocker,verminderten den basalen Efflux durch die basolaterale Membran im distalen Kolon. Den gleichen Effekt zeigte eine Versuchsserie mitBrefeldin A, welches den Transport von neu synthetisierten Proteinen aus dem Golgi-Apparat hemmt. Die Stimulation des Effluxes durchZellschwellung blieb jedoch unverändert. Im Gegenteil war die Stimulation des apikalen K+-Effluxes in Gegenwart von Inhibitoren desZytoskeletts wie Colchicin, welches eine Depolymerisation von Mikrotubuli auslöst, und von Brefeldin A sogar verstärkt. Die Versuche belegen, dass volumensensitive K+-Kanäle, die unter anderem durch Ca2+ und das Zytoskelett beeinflusst werden können,an der Regulation des transepithelialen K+-Transportes im Kolon beteiligt sind.de_DE
dc.description.abstractThe intestinal potassium transport is under the control of many second messenger systems (for example cAMP and Ca2+). Anotherpathway that influences Na+- and Cl--transport is the cell volume. Volume-activated K+-channels are present in the basolateral membrane ofcolonic crypts. Therefore the aim of this work was to investigate if there is a volume influence at the transepithelial K+ transport. The effect of cell swelling and cell shrinkage on K+ transport across the rat colonic epithelium was studied by measuring unidirectionalfluxes, uptake and efflux of 86Rb+, a marker for K+. Exposure to a hypotonic medium stimulated the secretory, serosa-to-mucosa flux of K+, whereas exposure to a hypertonic mediuminhibited the absorptive, mucosa-to-serosa flux of K+ in the distal, but not in the proximal colon. Neither manoeuvre had any effect on theuptake of K+ across the apical or the basolateral membrane. Cell swelling stimulated apical and basolateral K+ efflux in both colonic segments, whereas cell shrinkage reduced it. The stimulation of theswelling-induced K+ efflux in the distal colon is Ba2+ sensitive at the apical side and quinine sensitive at the basolateral side. Incubation ofthe tissue in Ca2+-free buffer and La3+, which blocks Ca2+-influx into the epithelium side, strongly reduced the basal K+ efflux across thebasolateral membrane. The same was observed with brefeldin A, a blocker of the transport of newly synthesized proteins out of theendoplasmatic reticulum. Swelling-induced K+ efflux, however, was not reduced. Hypotonically-stimulated apical K+ efflux was enhanced bydrugs disturbing the cytoskeleton such as colchicine, an inhibitor of the polymerisation of microtubules, and by brefeldin A. These results demonstrate that the cell volume is involved in the regulation of transepithelial K+ transport across the rat colonic epitheliumand suggest a role of Ca2+ and the cytoskeleton in the control of a part of the volume-sensitive K+ channels.en
