dc.description.abstractLäsionen des M. interosseus medius sind eine häufige Ursache für Lahmheiten und Verlust der Sporttauglichkeit des Pferdes. Die natürliche Heilung des Fesselträgers ist langsam und von schlechter Qualität, was zu langen Ausfällen führt und in einer Prädisposition für Rezidiverkrankungen resultiert. Für die Evaluierung der biomechanischen Eigenschaften einer kollagenen Struktur ist die Kenntnis über dessen Ultrastruktur essentiell. Diese Arbeit beschäftigt sich mit der Darstellung des M. interosseus medius auf ultrastruktureller Ebene, der Elektronenmikroskopie. Sie soll damit einen Beitrag zum Verständnis der Funktion und Heilung des Fesselträgers leisten. Außerdem dienen die Ergebnisse als Datengrundlage für den Vergleich von gesunden mit geschädigten Fesselträgern. Bisherige Studien haben ergeben, dass die Auswertung elektronenmikroskopischer Bilder eine gute Möglichkeit ist, um ein Verständnis über die Funktion unterschiedlicher Gewebe zu liefern. Es wurden insgesamt 4 Vorderbeine und 4 Hinterbeine von gesunden Pferden untersucht. An insgesamt 8 Lokalisationen wurden Proben des M. interosseus medius entnommen, um sie für die Elektronenmikroskopie vorzubereiten. Nach dem Einlegen in eine Fixierlösung (3% Glutaraldehyd/0,05ml Phosphatpuffer), wurden die Präparate nachfixiert (2% OsO4 und 0,2m Phosphatpuffer/Saccharose) und entwässert. Dann folgte die Einbettung in Kunstharz, die Herstellung des Semidünnschnitts, die Färbung mit Toluidin und der Ultradünnschnitt. Die Bilder wurden unter dem Transmissionselektronenmikroskop Philips CM 100 angefertigt. Nach dem Ausdrucken und Einscannen erfolgte die Auswertung mit dem Programm Image J. Folgende Parameter wurden berechnet: Fibrillenflächenanteil; Fibrillendichte; Fläche, Durchmesser und Umfang jeder Kollagenfibrille und die Fibrillenflächenverteilung. Die Ergebnisse zeigen einen Fibrillenflächenanteil am Vorderbein zwischen 54,5% und 62,2% und am Hinterbein zwischen 61,3% und 69,9%. Der Vergleich zeigt, dass an allen Lokalisationen am Hinterbein ein höherer Fibrillenflächenanteil als am Vorderbein gemessen wurde. Das bedeutet, dass der Fesselträger aufgrund seines höheren Anteils an Kollagenfibrillen stabiler und kräftiger sein müsste. Die Fibrillendichte beträgt am Vorderbein zwischen 94,6 und 163 Fibrillen pro Fläche (µm²) und am Hinterbein zwischen 64,6 und 145,4. Bei diesen Ergebnissen zeigt sich, je mehr kleine Fibrillen, desto größer die Fibrillendichte. Der Fibrillendurchmesser, sowie die Fibrillenfläche zeigen, dass am Hinterbein im Allgemeinen der Gehalt an dickeren Kollagenfibrillen höher ist als am Vorderbein. Der mediale Schenkel des Hinterbeins ist die Lokalisation mit dem höchsten Anteil dicker Fibrillen. An der Lokalisation Mitte lateral am Vorderbein wurden die meisten dünnen Fibrillen gemessen. Der Fibrillendurchmesser beträgt vorne zwischen 79,3nm und 101,3nm und hinten zwischen 84,1nm und 120,9nm. Dicke Fibrillen sind für die dehnbare Kraft einer Sehne (stress) und dünne Fibrillen dafür einer Überdehnung zu widerstehen (strain). Das bedeutet, dass der Fesselträger des Vorderbeins dehnbarer und der des Hinterbeins kräftiger ist. Der höhere Kollagenanteil am Hinterbein könnte eventuell durch das Protein COMP erklärt werden, welches für die Kollagenfibrillogenese von großer Bedeutung ist. Es ist möglich, dass im juvenilen Alter in der Hinterextremität mehr COMP vorhanden ist. Der höhere Anteil an dicken Fibrillen am Hinterbein könnte sich im Laufe der Zeit entwickelt haben, da Fesselträgerschäden hier häufiger auftreten und v.a. Dressurpferde vermehrt Last auf die Hinterbeine aufnehmen sollen. Das könnte eine Erklärung dafür sein, dass sich der ultrastrukturelle Aufbau des M. interosseus medius hinten im Vergleich zum Vorderbein verändert hat. Andererseits ist es ebenso denkbar, dass die vermehrte Belastung der Vorhand zu einer Spaltung großer Fibrillen führt. Das würde in einem höheren Anteil kleiner Fibrillen resultieren. Die unterschiedliche Verteilung der Kollagenfibrillen könnte die Therapiewahl beeinflussen, indem am Hinterbein die Behandlung mit PSGAG vorgezogen würde und am Vorderbein die mit Hyaluronsäure.Die Arbeit hat einen ersten Einblick in den ultrastrukturellen Aufbau des Fesselträgers ermöglicht. Sie soll einerseits ein Anstoss für weitere elektronenmikroskopische Untersuchungen geben, um den Aufbau des M. interosseus medius noch besser zu verstehen und andererseits eine Vergleichsgrundlage für die ELMI-Bilder geschädigter Fesselträger sein.de_DE
dc.description.abstractLesions of the M. interosseous medius are a frequent reason for lameness and loss of suitability of sports of the horse. The natural healing of the suspensory ligament is slow and of bad quality, what leads to long failures and results in a predisposition for recurrent diseases. The knowledge of the ultrastructure is essential for the evaluation of the biomechanic properties of the collagenous structure. This dissertation is concerned with the presentation of the M. interosseous medius on the ultrastructural level, the electron microscopy. The dissertation should be helpful as a contribution of comprehension for the function and the healing of the suspensory ligament. Furthermore the results serve as a data base for the comparison of a healthy and a damaged suspensory ligament. Past Studies have shown that the evaluation of electron microscope pictures are a good possibility to get a better understanding of the different tissues. 4 front legs and 4 hind legs of healthy horses were examined. On 8 localisations of the M. interosseous medius samples were abstracted to prepare them for the electron microscope. After inserting the samples in a fixer (3% glutaraldehyde/0,05ml phosphate buffer), the anatomical preparations were fixed again (2% OsO4 and 0,2m phosphate buffer/saccharose) and dehydrated. After that the preparations were embedded in synthetic resin, the preparation of the semi thin section, the colourisation with Toluidin and the ultra thin section. The pictures were taken under the transmission electron microscope Philipps CM 100. The evaluation with the software Image J followed after printing and scanning the pictures. The following parameters were calculated: fibril area fraction; fibril density; area, diameter and scope of each collagen fibril and the fibril area distribution. The results show that the fibril area fraction in the forelimb is between 54.5% and 62.2% and in the hindlimb between 61.3% and 69.9%. The comparison shows that on each localisation of the hindlimb the fibril area fraction is higher than measured on the forelimb. This means that the suspensory ligament because of its higher proportion on collagen fibrils must be more stable and powerful. The fibril density on the forelimb is between 94.6 and 163 fibrils per area (ym²) and on the hindlimb between 64.6 and 145.4. The results show that in case of more smaller fibrils the fibril density is bigger. The fibril diameter as well as the fibril area show that the concentration of thicker collagen fibrils on the hindlimb are higher than on the forelimb. The medial branche of the hindlimb is the location with the highest amount of thick fibrils. On the localisation middle lateral most of the thin fibrils were measured. The fibril diameter in the forelimb is between 79.3nm and 101.3nm and on the hindlimb between 84.1nm and 120.9nm. The thick fibrils are for the tensile strength of a tendon (stress) and the thin fibrils to withstand a hyperextension (strain). This means that the suspensory ligament of the forelimb is more tensile and the one of the hindlimb is more powerful. The protein COMP could be a reason for the higher amount of collagen on the hindlimb and is also important for the collagen fibrillogenesis . It seems to be possible that in the juvenile age the hindlimb contained more COMP. The higher amount of thick fibrils in the hindlimb could be a part of an evolution process, as damages on suspensory ligament are more frequently because dressage horses take more load on the hind legs. This could be a reason for the fact that the ultrastructural constitution of the M. interosseous medius on the hind in comparison to the front leg has changed. On the other hand it is also thinkable that the increased load on the front hand caused a split of the fibrils. The consequence of this would be a bigger part of small fibrils. The different distribution of the collagen fibrils could influence the therapy, by using PSGAG for the treatment on the hindlimb and hyaluronic acid for the forelimb. This dissertation provides an insight into the ultrastructural constitution of the suspensory ligament. On the one hand this dissertation should initiate more electron microscope studies for a better understanding of the structure of the M. interosseous medius and on the other hand as a basis of comparison for electrone microscope pictures of damaged suspensory ligament.en
