dc.description.abstractGRAF - möglicherweise ein Tumorsupressorgen für MDS/AML? Die häufigste strukturelle Veränderung bei MDS und AML ist die Deletion des langen Arms des Chromosoms 5. Obwohl sowohl die Größe als auch die Lage der deletierten Segmente erheblich variieren kann, so ist dennoch in der Mehrheit der Fälle die Bande 5q31 verloren. Diese Erkenntnis führte zu dem Schluss, dass diese kritische Region ein Tumorsupressorgen beherbergen muss, dessen Verlust oder Inaktivierung eine entscheidende Rolle bei der Entstehung maligner myeloischer Erkrankungen spielt. Die Variabilität der Deletionen und die große Anzahl der möglicherweise in Frage kommenden Gene innerhalb dieser Region haben die Identifikation und Isolation solch eines Gens erheblich erschwert. Durch die molekulargenetische Untersuchung von Knochenmarks- bzw. Blutproben von 14 MDS/AML Patienten, die in der zytogenetischen Unter-suchung eine Deletion 5q aufwiesen, sollte in Anlehnung an die two-hit-Hypothese von Knudson gezeigt werden, dass ein Ausfall beider Allele des GRAF-Gens bei Patienten mit MDS/AML vorkommt. Dabei kann es sich entweder um eine größere Deletion oder eine Punktmutation handeln. Für die Hypothese, dass es sich bei GRAF um ein Tumorsupressorgen handelt, spricht die in der Literatur beschriebene Funktion von Graf, Rho, welches als Stimulator der Ras vermittelten DNA-Synthese angesehen wird, zu deaktivieren. Auch die Tatsache, dass wir bei 8 der 14 untersuchten Patienten, die eine Deletion eines Allels aufwiesen, funktionell wichtige Sequenzveränderungen in dem verbleibenden Allel gefunden haben, lässt vermuten, dass Deletionen und Mutationen des GRAF-Gens eine entscheidende Rolle bei der Entstehung und dem Verlauf hämatologischer Erkrankungen spielen und lässt auf einen möglicherweise tumorunterdrückenden Effekt von GRAF schließen. Es werden jedoch noch weitere Experimente nötig sein, um die Beziehung zwischen RhoA, p21 und GRAF im Zusammenhang mit der Entwicklung von Leukämien und möglicherweise anderer Tumoren klären zu können. Aufgrund der Tatsache, dass die Mehrzahl der von uns untersuchten Patienten die GRAF-Wildtypsequenz in dem verbliebenen Allel aufwiesen, kamen wir zu dem Schluss, dass neben GRAF auch der Verlust anderer Gene bei der Entwicklung von Leukämien mit Deletion 5q eine Rolle spielen muss.de_DE
dc.description.abstractGRAF probably a tumor supressor gene of AML/MDS The deletion of the long arm of chromosome 5 is the most frequent structural abnormality in MDS and AML. Although the size and position of the deleted segment may vary considerably, band 5q31 is lost in most cases. This finding led to the notion that the critical region must harbor a tumor supressor gene whose loss or inactivation is critical for the development of these malignant myeloid disorders. The variability of the deletions and the large number of attractive candidate genes within this region has hampered the identification and isolation os such a gene. By molecular analysis of blood and bone marrow samples in 14 MDS/AML patients with a deletion 5q it was to be shown, in reference to the two-hit hypothesis of Knudson, that a loss of both allels of the GRAF (GTPase regulator assosiated with the focal adhesion kinase pp125 FAK) gene is found in MDS/AML patients, which can either be a deletion or a point mutation. The chicken Graf gene, which is to 90% homologous to the human GRAF gene, acts as a negative regulator of Rho. Olson ad al found recently that Rho suppresses p21 41, a well-known inhibitor of the cell cycle. The loss of function of Graf prevents the physiologic down-regulation of Rho. Thus, the mitigation or elimination of the negative regulatory function of Graf may lead to the repression of p21. This observation leads to the hypothesis that GRAF may be a tumor supressor gene. In 8 of 14 patients with a deletion of one allel we found functional important changes of the sequence in the remaining allel. This leads to the supposition that deletions and mutations in the GRAF gene play an important role in the development and course of haematological diseases and that GRAF may have a tumor supressor function. Further experiments are needed to clarify the relationship between RhoA, p21 and GRAF in the context of development of leukemias and probably other malignant diseases. However, based on the fact that most of our patients displayed the GRAF wild-typ sequences within the second GRAF allel we conclud that apart from GRAF also the loss of other genes play an important role in the development of leukemias with deletion 5q.en
