dc.description.abstractIn dieser Arbeit wurde erstmals in einem Tiermodell mit durch Endothelin induziertem cerebrovaskulären Insult die Differenzierung und funktionelle Integration von neuronalen xenotransplantierten eGFP markierten embryonalen Stammzellen mit Hilfe der patch-clamp Technik und immunhistochemischen Färbungen untersucht. · In etwa 30% von eGFP positiven Zellen konnten Einwärts- und Auswärtsströme nachgewiesen werden. · Die transplantierten Zellen überlebten im elektrophysiologischen Beobachtungszeitraum von 7 Wochen nach Transplantation. · Die Einwärtsströme, die eine für spannungsabhängige Natriumkanäle typische Kinetik zeigten, konnten durch 100 nM TTX blockiert werden. · Nur diese Zellen konnten Aktionspotentiale generieren und wurden deshalb neuronen-ähnliche Zellen genannt (neuron-like cells: NLC)· Die Auswärtsströme zeigten eine langsame Inaktivierungskinetík (τ>500 ms) und konnten durch 10 mM TEA auf 20 % blockiert werden. · Die Auswärtsströme gliedern sich in zwei Komponenten Kdr (delayed rectifier K+-Kanäle) und KCa (calciumabhängige K+-Kanäle). · Kdr hat großen Einfluss auf das Ruhepotential der untersuchten NLCs. · KCa führt zu einer Modulation der Aktionspotential Dauer.· Mit zunehmender Zeit nach Transplantation wurde das Ruhemembranpotential zunehmend negativer. · Die Reifung der untersuchten Zellen führte zu einer Verkürzung der Aktionspotential Dauer und zu der Fähigkeit, Serien von Aktionspotentialen zu generieren. · In NLCs konnten spontane Einwärtsströme von kurzer Dauer und mit ähnlicher Kinetik beobachtet werden, die als exzitatorische postsynaptische Potentiale (EPSPs) identifiziert wurden. · Die EPSPs sind ein Hinweis auf die funktionelle Integration der NLCs. Offen bleibt, ob die Verknüpfung untereinander oder mit host-Neuronen ausgebildet wird. · Die calciumabhängigen EPSPs führten zum spontanen Feuern von Aktionspotentialen. · Die aus transplantierten Zellen differenzierten Gliazellen besaßen nur Auswärtsströme mit geringerer Amplitude und konnten keine Aktionspoteniale generieren.de_DE
dc.description.abstractFor the first time, the differentiation and functional integration of xenotransplanted eGFP+ stem-cells derived from neuronal embryonic precursor-cells was examined in an animal, endothelin-induced stroke model. · Inward and outward currents were found in about 30 % of eGFP positive cells. · Transplanted cells survived during the electrophysiological observation period of 7 weeks after transplantation. · The inward currents, which demonstrated the typical kinetics of voltage-gated sodium currents were blocked by 100 nm TTX. · Only the eGFP+-cells were able to generate action potentials. Therefore they were called neuron-like cells (NLC). · The outward current revealed slow inactivation kinetics (τ>500 ms) and 10 mM TEA could suppress it to 20%. · The outward currents consisted of two components, Kdr (delayed rectifier K+-currents) and KCa (calcium dependent K+-currents). · Kdr had a large influence on the resting membrane potential of NLC. · KCa modulated the actionpotential duration. · The resting membrane potential became more negative during the electrophysiological observation period. · As the cells matured, the action potential duration was shortened and they were more able to generate trains of action potentials. · Spontaneous inward currents of short duration and similar kinetics were observed in NLCs and identified as excitatory postsynaptic potentials (EPSPs). · The EPSPs are evidence of the functional integration of NLC. It is unclear if NLC interconnect with each other or with the host-neurones.· The calcium-dependent EPSPs evoked spontaneous firing of actionpotentials in NLCs. · Differentiated glial cells derived from transplanted cells possessed only voltage-dependent outward currents with low amplitude and could not generate an action potential.en
