dc.description.abstractDas PTPIP 51, ein möglicher Interaktionspartner der PTP1B und TCPTP, findet sich bei vielen Säugetieren in unterschiedlichsten Geweben sowie in verschiedensten Zelllinien. Eine Assoziation zu proliferierenden Zellen weist auf die Beteiligung des PTPIP51 an Proliferationsvorgängen hin. Auf Grund des Verteilungsmusters von PTPIP51 in der Epidermis kann von einer Assoziation zu Differenzierungsprozessen ausgegangen werden. Eine Überexpression in Zellkultur führte zur Apoptose.Ziel dieser Arbeit war es, mögliche Zuordnungen zu diesen Funktionen während der Embryonalentwicklung der Maus zu untersuchen.Mittels Immunoblotting, RT-PCR, Immunhistochemie und in situ Hybridisierung konnte das PTPIP51 in allen untersuchten Geweben während aller Stadien nachgewiesen werden. Die Expression des PTPIP51 erfolgte in Abhängigkeit vom Entwicklungsstadium. Detailliert untersucht wurden: die Entwicklung der Augen, des Plexus choroideus, der Zunge und der Lunge.Im Immunoblotting wurde die Expression verschiedener Isoformen nachgewiesen. Die Molekulargewichte der gefundenen Isoformen stimmen mit den theoretisch berechneten Größen überein.Während der Augenentwicklung war das PTPIP51 Antigen zu allen untersuchten Stadien in suprabasalen Zelllagen der Epidermis nachweisbar. In Conjunctiva und Cornea gelang der Nachweis bei allen untersuchten Stadien. Während der Cornea-Entwicklung kam es zur Expression im Cornealepithel, dem Endothel und ab 16. dpc im Stroma. Diese PTPIP51 reaktiven Strukturen behalten ihre Fähigkeit zur weiteren Differenzierung bei: das Epithel der Cornea und der Conjunctiva können sich in verhornendes Plattenepithel umwandeln, bei Traumen kommt es ausgehend von den genannten Strukturen Reparaturprozessen.Während der Linsenentwicklung war das untersuchte Antigen in allen untersuchten Stadien im anterioren Linsenepithel nachweisbar. Auch hier bleibt die Differenzierungfähigkeit zeitlebens erhalten.In der sich entwickelnden Neuroretina war PTPIP51 ab 12. dpc nachweisbar. In frühen Stadien war die innere Neuroblastenschicht und die sich entwickelnde Rezeptorzellschicht reaktiv. Während der weiteren Entwicklung kam es zu einer Expression in der inneren Neuroblastenschicht, hier vor allem in den zukünftigen Ganglienzellen, in der inneren plexiformen Schicht, der äußeren plexiformen Schicht und der Rezeptorzellschicht. Beim adulten Auge verbleiben genannte Strukutren bis auf die äußeren Neuroblastenschicht PTPIP 51 positiv.In vivo konnte keine Assoziation des Proteins zur Apoptose während der Augenentwicklung festgestellt werden. Kolokalisationsstudien mit Synaptophysin und Calbindin machen eine Funktion von PTPIP51 bei der Ausbildung von Synapsen wahrscheinlich.In allen untersuchten Stadien war die EOM stets PTPIP51 reaktiv. Auch hier besteht eine Assoziation zur Differenzierung, da die Muskeln stets embryonales Myosin exprimieren und einer kontinuierlichen Differenzierung unterliegen.Ab 12. dpc fiel eine starke Expression im Plexus choroideus in apikalen- und basalen Anteilen des Plexusepithels auf. Die Plexus choroidei spielen von Beginn an eine essentielle Rolle für die Entwicklung des ZNS. Eine mögliche Assoziation zu den spezifischen Zell-Zell-Kontakten und/oder den Transportsystemen ist wahrscheinlich. Auch das Plexusepithel behält die Fähigkeit zur Differenzierung bei. Es hat z.B. bei Reparaturmechanismen im Rahmen hypoxischer Schädigungen eine entscheidende Bedeutung.In dem Epithel des zukünftigen Mundbodens, des Gaumens und der Zunge ist ab 12. dpc eine Expression detektierbar. Ab 16. dpc, zeigen sich das Epithel des Mundbodens und der Zunge reaktiv. Hier, aber auch während der Entwicklung aller anderen untersuchten Organe, kommt es zur Interaktion von Geweben mit Ursprung in den drei Keimblättern des Embryos. Diese Interaktionen werden über verschiedene Wachstumsfaktoren reguliert. In den zugehörigen Signalkaskaden finden sich Tyrosinphosphorylierungen und Dephosphorylierungen an deren Übermittlung PTPIP51 ggf. beteiligt ist. Auch bei der Lungenentwicklung spielen diese Mechanismen eine wichtige Rolle.Hier ließ sich ab 12. dpc eine Reaktion mit dem gegen das PTPIP51 gerichtete Antigen im respiratorischen Epithel der Haupt- und Segmentbronchien sowie im umgebenden Mesenchym nachweisen. Das Lungengewebe ist ebenfalls durch Zell-Zell-Kontakte und einen Oberflächenbesatz mit Kinocilien charakterisiert, für deren Ausbildung und Funktion PTPIP51 möglicherweise mit verantwortlich ist.Im Bereich der Lunge kommt es bei der Metaplasie zu Plattenepithel oder bei der Umwandlung zu Becherzellen und auch bei der Reparatur von traumatischen Defekten zu Differenzierungsprozessen innerhalb des respiratorischen Epithels ausgehend von den PTPIP51 positiven Zellen.de_DE
dc.description.abstractProtein tyrosine phosphatase interacting protein 51 a potential interacting partner of two highly homologous phosphatases, namely PTP1B and TCPTP, was found in various mammalian tissues as well as in different cultured cell lines. Its association to proliferating cells indicates a possible role of PTPIP51 in proliferation processes. Due to the specific expression of PTPIP51 in the epidermis an association of PTPIP51 to differentiation processes is likely. An overexpression of PTPIP51 in cultured cells results in apoptosis.Aim of this study was to investigate, whether the protein is associated to the above mentioned processes in embryonic development of the mouse. During embryogenesis a balanced interplay between proliferation, differentiation and apoptosis is required for organogenesis and therefore is essential for all developmental processes.The expression of PTPIP51 during mouse development was studied by RT-PCR, in situ hybridization, immunoblotting and immunohistochemistry. The expression of PTPIP51 could be demonstrated in all investigated stages. Moreover, the quantity of PTPIP51 expression was shown to be dependent on the developmental stage. A more detailed analysis was performed for the eye, the choroid plexus, the tongue and lung tissue.Immunoblotting experiments suggest the development and tissue dependent expression of different putative isoforms, whose molecular weights match with calculated theoretical data. This holds particularly true for the postpartal development of mouse eye.During eye development PTPIP51 protein was found in suprabasal layers of the epidermis. Regarding corneal tissue, PTPIP51 could be traced in the corneal epithelium and endothelium during all investigated stages. Moreover, from day 16 pc onwards keratocytes also expressed PTPIP51 protein. From day 12 pc unto the adult stage, PTPIP51 was found in the anterior lens epithelium.It is noteworthy that all PTPIP51-positive structures keep their life-long potential for differentiation, the later being important in repair processes following trauma.The evolving neuroretina displayed PTPIP51 protein in the inner neuroblastic as well as in the developing photoreceptor cell layer. Ongoing development led to an enhanced expression of PTPIP51 in both layers. In addition, PTPIP51 was upregulated in (the formerly negative) inner and outer plexiform layer. PTPIP51 expression persists in the adult stage except for the outer neuroblastic layer.Apoptotic processes play a central role during eye development, yet, an in vivo association of TUNEL-labelled cells with PTPIP51 expression could not be observed.Since immunolabeling studies with PTPIP51 and synaptophysin, as well as calbindin revealed a partial colocalization, one may speculate about a role of PTPIP51 in synapsis formation.EOM were PTPIP51-positive during all investigated stages. These muscles express embryonic myosin and undergo a constant redifferentiation. Therefore, PTPIP51 may serve as a signalling partner in this process.On day 12 pc the apical and basal part of cells comprising the choroid plexus were PTPIP51-positive. The apical cell pole showed PTPIP51-immunreactive granula, which may correspond to the basal bodies of kinocilia.The choroid plexus is essentially required for CNS development harbouring the blood- brain barrier, which is characterized by tissue-specific cell-cell contacts.The choroid epithelium retains its ability to differentiate and plays therefore an important role for regenerating processes following hypoxic damage.On day 12 pc, PTPIP51 protein could be traced in the epithelium of the developing floor of the oral cavity, palate and tongue. From E16 onwards PTPIP51-labeling showed a steadily stronger reaction with the PTPIP51-antibody.Tongue formation in particular but also the development of all other investigated organs ultimately evolving from the three germ layers are regulated by growth factors and their corresponding receptors and tyrosine-phosphorylation dependent signalling cascades. PTPIP51 may serve as a signalling partner in these cascades.The respiratory epithelium of the main- and segmental bronchi of the lung bud were found to be PTPIP51-positive. Some surrounding stromal cells also expressed PTPIP51. Since this epithelium harbours kinocilia as well as specific cell-cell contacts, one may hypothesize that PTPIP51 is mandatory for signalling pathways associated with cell-cell interaction and animation of kinocilia. Metaplasia to squamous epithelium, as well as reparation events require cells, which retain the ability to differentiate. As stated above, PTPIP51 expression particularly occurs in these tissues and therefore may be important for these processes.en
