dc.description.abstractDiese Arbeit diente der Methodenoptimierung zur Identifizierung exkretorisch-sekretorischer Proteine des Nematoden C. elegans sowie der Untersuchung der Effekte von RNA-Interferenz knock-downs der drei ausgewählten Proteine GalNAc-Transferase GLY-10 sowie der Aspartylproteasen ASP-2 und ASP-6 auf die Zusammensetzung des Sekretoms.Die Proteine aus dem Sektretom von C. elegans wurden durch ein- und zweidimensionale Gelelektrophorese aufgetrennt. Nach in-gel Verdau mit Trypsin wurden die Proteine mittels Massenspektrometrie identifiziert. Durch die Methodenoptimierung bei der Probenaufreinigung konnte die Zahl der identifizierbaren Proteine deutlich gesteigert werden.Zusammengefasst lässt sich sagen, dass nach den RNAi knock-downs eine geringere Anzahl an Proteinen identifiziert werden konnte als in den Sekretomen aus dem ersten Teil der Arbeit oder dem Sekretom des leeren Kontrollvektors. Es zeigte sich auch eine deutliche Verschiebung innerhalb der funktionellen Proteinklassen. Es dominierten Struktur- und Stoffwechselproteine, während vor allem nach dem ASP-6- und GalNAcT knock-down hauptsächlich Proteine mit bislang unbekannter Funktion identifiziert wurden. Bemerkenswert war die Identifikation von Chaperonen nach GalNAcT knock- down , welche in keinem anderen Experiment identifiziert werden konnten.Die Ergebnisse zeigen, dass die untersuchten Aspartylproteasen und die GalNAc-Transferase große Bedeutung für die Zusammensetzung des Sekretoms haben. Daraus lässt sich die Hypothese ableiten, dass die Verwendung von Protease- oder Glykosylierungsinhibitoren ein interessanter und vielversprechender Ansatzpunkt für die Entwicklung neuer Bekämpfungsstrategien hinsichtlich parasitärer Erkrankungen darstellt, da hiermit evtl. das immunmodulatorische Potential der Sekretome signifikant verändert und die Überlebensfähigkeit parasitärer Nematoden herabgesetzt werden könnte.de_DE
dc.description.abstractThis thesis served for the optimization of methods for the identification of excretory- secretory proteins from the nematode C. elegans and the investigation of the changes within the secretom protein composition after RNA interference targeting three proteins: the GalNAc-transferase GLY-10, the aspartylprotease 2 and the aspartylprotease 6.The secretome proteins of C. elegans were separated by one- and two-dimensional gel electrophoresis. After in-gel digestion with trypsin, the proteins were identified by mass spectrometry. The method optimization led to a significant increase of the number of identified proteins.Summarizing, the thesis showed a reduced number of identified proteins after RNA interference knock-down experiments compared to the secretome of the first part of the thesis and the secretome of the control vector. There was a significant change within the functional classes of the identified proteins observed. Structural and metabolic proteins dominated the secretomes, whereas especially after ASP-6 and GalNAcT knock-down proteins with unknown function were highly abundant. The most apparent difference after GalNAcT knock-down was the identification of chaperons, which were not identified in the other RNAi experiments.The results show that the aspartylproteases and the GalNAc-transferase have significant impact on the composition of the secretomes. This raises the hypothesis that the use of protease- or glycosylation inhibitors might be an interesting and promising approach for the development of new anthelminthic strategies thus changing the immunomodulatory potential of the secretome significantly and lowering the survival of parasitic nematodes.en
