In vitro Expression von Integrinen und extrazellulärer Matrix in bovinen Plazentazellen

Abstract

Integrine, eine Familie heterodimerer Transmembranrezeptoren, sind nicht nur für das Anheften von Zellen an ihre Liganden der extrazellulären Matrix (ECM) verantwortlich, sondern auch für die Transduktion von Signalen. Durch die so genannte eingeschränkte Trophoblastinvasion bietet die epitheliochoriale Rinderplazenta gute Bedingungen, um invasive Prozesse zu untersuchen. Unsere Arbeitsgruppe zeigte bereits, dass die Migration der TGC im bovinen Plazentom von der Expression spezifischer Integrine und ihrer ECM Liganden begleitet wird. Vor diesem Hintergrund wurden in der vorliegenden Arbeit Primärkulturen verschiedener Zellpopulationen aus dem Rinderplazentom etabliert und charakterisiert und mit den in vivo Verhältnissen verglichen. Hierfür wurde die Methode zur Zellgewinnung und -kultivierung für das Rinderplazentom adaptiert und optimiert. Due to the phenomenon of restricted trophoblast invasion, the bovine epitheliochorial placenta offers intriguing conditions to study invasive processes. Our group has previously shown that the migration of TGC in the bovine placentome is accompanied by the expression of specific integrins and their corresponding ECM ligands. On the base of these studies, primary cultures of different cell populations from cow placentomes were established, characterized and compared with in vivo conditions. As a precondition the method for cell isolation and cultivation had to be adapted and optimized for bovine placentomal cells.
