Item The effect of dry cow antibiotic with and without an internal teat sealant on udder health during the first 100 d of lactation : a field study with matched pairs(2012) Mütze, Katja; Wolter, Wilfried; Failing, Klaus; Kloppert, Bärbel; Bernhardt, Heinz; Zschöck, MichaelThe objective of this field study was to compare the udder health status as well as the clinical mastitis rate during the first 100 d of lactation in cows that received long-acting dry cow antibiotic alone (group AB) or in combination with an internal teat sealant (group AB + OS). The study was conducted during a 9-month period and included 136 Holstein cows from 12 dairy farms in Hessia, Germany. Between days 1 and 5 after calving a California mastitis test (CMT) was performed. Milk-samples were collected for bacteriological culture before drying off, between days 6 and 14 and days 35 and 56 of lactation. Additionally the cows were monitored for the occurrence of clinical mastitis events until 100 d post partum. Within the 12 herds cow-pairs were formed on the basis of age, milk yield and SCC. A cow-pair consisted of one cow from group AB and one cow from group AB + OS. For statistical analysis within every cow-pair one quarter that has been dried off with internal teat sealant and dry cow antibiotic (group AB + OS) was compared with one quarter that has been dried off with dry cow antibiotic (group AB) alone. As criterion for the matching process of udder quarters the cytobacteriological udder health status before drying off was used. A total of 544 quarters (136 cows) were used in this analysis. In the first 5 d after calving, group AB had significantly more quarters with a positive CMT reaction than group AB + OS (85 vs. 57; P <0·001), and in the first 100 d of lactation, group AB had more quarters with clinical mastitis than group AB + OS (25 vs. 15; P = 0·03). In the time periods 6 14 and 35 56 d of lactation, there were fewer quarters in group AB + OS populated with Corynebacterium spp. (days 6 14, P = 0·05; days 35 56, P = 0·02) and aesculin-positive streptococci (days 35 56, P = 0·02). The internal teat sealant was a promising tool for the prevention of new intramammary infections (IMI) of dry cows with environmental udder pathogens as expressed during early lactation.Item Influence of selected (pre-)maturational parameters on in vitro development and sex distribution of bovine embryos(2012) Abele, Eva; Stinshoff, Hanna; Hanstedt, Ana; Wilkening, Sandra; Meinecke-Tillmann, S.; Wrenzycki, ChristineThe objectives of this research were to study the influence of a reduced oxygen concentration during in vitro maturation (IVM) and examine the effect of follicular glucose concentration on bovine in vitro development and sex distribution. In the first experiment, abattoir-derived cumulus oocyte complexes (COC) were matured under 5% O2 or 20% O2. Secondly, COC were isolated and the glucose (G) concentration of each follicle was determined. COC were pooled in groups (G<1.1mMol or G=1.1mMol) according to the glucose content before being subjected to in vitro production (IVP). Cleavage and development rates were assessed on days 3, 7 and 8 post insemination. Blastocysts of each group were sexed by polymerase chain reaction (PCR). Expanded blastocysts were stained to assess total cell numbers and live dead cell ratio. Cleavage and development rates stayed similar after reducing the O2 concentration during IVM. The sex ratio of embryos generated from oocytes matured under 5% O2 was shifted in favour of the female (?: 61.9%), whereas the sex ratio of embryos belonging to the IVM 20% O2 group did not differ significantly from the expected 50:50 ratio. Neither a higher nor a lower intrafollicular glucose concentration influenced cleavage and development rates, cell numbers or live dead cell ratio. Eighty five per cent (G<1.1) and 63.6% (G=1.1) of the analysed embryos were female. In summary, neither a reduced O2 concentration during IVM nor selection based on follicular glucose concentrations affected the morphological quality of embryos. Although the sex distribution was shifted in favour of female embryos in all three experimental groups, more male embryos could be seen in the G=1.1 group compared with the G<1.1 group.Item Metastasizing esthesioneuroblastoma in a dog(2015) Siudak, Krystyna; Klingler, Melanie; Schmidt, Martin J.; Herden, ChristianeA 7-year-old Afghan hound presented with a history of disorientation, loss of vision, and seizures. Magnetic resonance imaging helped identify a mass at the level of the main olfactory bulb that compressed and displaced adjacent tissues in the cribriform plate into the nasal cavity and nasopharynx. Bony structures were osteolytic. After removing almost 80% of the mass, the tumor recurred a few months later. Due to severe respiratory distress and subsequent to an ultrasound diagnosis of a liver tumor, the dog was euthanized. In addition to the nasal mass, a single nodule in the liver and multiple nodules in the lung were present. All masses had similar cell morphology and were diagnosed as metastasizing esthesioneuroblastoma. The neoplastic cells expressed neuron-specific enolase and chromogranin A, and a few cells within the nasal mass were positive for cytokeratin. This is the first description of a canine esthesioneuroblastoma with distant metastases.Item Parasites of the grouper fish Epinephelus coioides (Serranidae) as potential environmental indicators in Indonesian coastal ecosystems(2015) Kleinertz, Sonja; Palm, H.W.A total of 195 Epinephelus coioides (Hamilton, 1822) were studied for fish parasites from Javanese (Segara Anakan lagoon) and Balinese waters. Up to 25 different parasite species belonging to the following taxa: one Ciliata, one Microsporea, five Digenea, one Monogenea, four Cestoda, four Nematoda, one Acanthocephala, one Hirudinea and seven Crustacea were identified with four new host and locality records. The dominant parasites included the monogenean Pseudorhabdosynochus lantauensis (53.3-97.1%), the nematode Spirophilometra endangae (23.3-42.9%), the digenean Didymodiclinus sp. (2.9-40.0%), the nematodes Philometra sp. (22.6-34.3%) and Raphidascaris sp. (2.9-28.6%), and the isopod Alcirona sp. (6.7-31.4%). Regional differences for E. coioides were found in terms of endoparasite diversity, total diversity according to Shannon Wiener, Simpson index and Evenness. A comparison with published data from Sumatera revealed highest endoparasite diversity (Shannon Wiener: 1.86/1.67-2.04) and lowest ectoparasite/endoparasite ratio (0.73/0.57-0.88) off the Balinese coast, followed by Lampung Bay, Sumatera (1.84; 0.67), off the coast of Segara Anakan lagoon (1.71; 0.71), and in the lagoon (0.30/0.19-0.66; 0.85/0.67-1.00). The presented data demonstrate the natural range of these parameters and parasite prevalences according to habitat and region, allowing adjustment of the scale that has been used in the visual integration of the parasite parameters into a star graph. The parasite fauna of E. coioides in Segara Anakan lagoon ´improved´ from 2004 until 2008/09, possibly related to earlier oil spill events in 2002 and 2004. The use of grouper fish parasites as an early warning system for environmental change in Indonesian coastal ecosystems is discussed.Item Paving the way for transgenic schistosomes(2012) Beckmann, S.; Grevelding, C. G.In parasitological research, significant progress has been made with respect to genomics and transcriptomics but transgenic systems for functional gene analyses are mainly restricted to the protozoan field. Gene insertion and knockout strategies can be applied to parasitic protozoa as well as gene silencing by RNA interference (RNAi). By contrast, research on parasitic helminthes still lags behind. Along with the major advances in genome and transcriptome analyses e.g. for schistosomes, methods for the functional characterization of genes of interest are still in their initial phase and have to be elaborated now, at the beginning of the post-genomic era. In this review we will summarize attempts made in the last decade regarding the establishment of protocols to transiently and stably transform or transfect schistosomes. Besides approaches using particle bombardment, electroporation or virus-based infection strateies to introduce DNA constructs into adult and larval schistosome stages to express reporter genes, first approaches have also been made in establishing protocols based on soaking, lipofection, and/or electroporation for RNA interference to silence gene activity. Although in these cases remarkable progress can be seen, the schistosome community eagerly awaits major breakthroughs especially with respect to stable transformation, but also for silencing or knock-down strategies for every schistosome gene of interest.Item Therapeutic targeting of glutaminolysis as an essential strategy to combat cancer(2019-05-22) Matés, José M.; Di Paola, Floriana J.; Campos-Sandoval, José A.; Mazurek, Sybille; Márquez, JavierMetabolic reprogramming in cancer targets glutamine metabolism as a key mechanism to provide energy, biosynthetic precursors and redox requirements to allow the massive proliferation of tumor cells. Glutamine is also a signaling molecule involved in essential pathways regulated by oncogenes and tumor suppressor factors. Glutaminase isoenzymes are critical proteins to control glutaminolysis, a key metabolic pathway for cell proliferation and survival that directs neoplasms’ fate. Adaptive glutamine metabolism can be altered by different metabolic therapies, including the use of specific allosteric inhibitors of glutaminase that can evoke synergistic effects for the therapy of cancer patients. We also review other clinical applications of in vivo assessment of glutaminolysis by metabolomic approaches, including diagnosis and monitoring of cancer.
