dc.description.abstractModifikationen der genregulatorischen Elemente des Fascinpromotors zur Entwicklung einer DNA-Vakzine stellen die aussichtsreichste Perspektive dar, eine starke, auf DC begrenzte Expression des codierten Proteins zu erreichen. Eine Duplikation unterschiedlich großer Bereiche der distal lokalisierten Homologiebereiche A und B liefert vornehmlich für jene der Homologieregion B eine robuste Verstärkung der Expression in Dendritischen Zellen, deren Spezifität auf diese Zellen erhalten bleibt. Keratinozyten, die bei einem Einsatz als DNA-Vakzine unter Verwendung einer Genpistole überwiegend am Applikationsort vorliegen, zeigen für die Duplikationskonstrukte nur eine geringe, basale Promotoraktivität. Die Untersuchung einiger dieser Konstrukte in der murinen DC-Zell-linie XS106 liefert eine ähnlich große Verstärkung der Expression wie in humanen DC und ermöglicht somit auch einen potentiellen Einsatz dieser DNA-Vakzine im Mausmodell. Durch Einführen einer repetitiven konsensus NFKappaB-Bindungsstelle an inerter Stelle im Expressionsplasmid, mit dem Ziel, den Kerntransport dieses Plasmids zu forcieren, kann zwar auch eine deutliche Verstärkung der Expression eines nachgeschalteten Reportergens erhalten werden, gleichzeitig geht dabei aber die Spezifität auf Dendritische Zellen verloren. Eine Fascin-Promotor eigene, repetitive NFKappaB-Bindungsstelle, an gleicher Stelle ins Plasmid kloniert, liefert keinen verstärkenden Effekt und ist somit zur Optimierung der Expressionseffizienz ebenfalls ungeeignet. In der vorliegenden Arbeit konnte auch gezeigt werden, dass im Modellsystem der SP37A3-Zellen, einer zur Ausreifung induzierbaren, murinen DC-Zell-linie, durch spezifische Inhibition des NFkappaB-Signalweges bzw- der MAP-Kinase p38 mit pharmakologischen Inhibitoren Dendritische Zellen resultieren, die eine verminderte Expression der zentralen kostimulatorischen Moleküle der B7-Gruppe, CD80 und CD86, aufweisen. Für SB203580, dem Inhibitor der MAP-K p38, konnte zusätzlich auch eine verminderte Expression weiterer Oberflächenrezeptoren dieser Gruppe, von CD273 bzw. CD274, nachgewiesen werden. Die reduzierte Expression dieser kostimulatorischen Moleküle lässt eine andere Qualität der Wechselwirkung mit T-Zellen vermuten, so dass durch Intervention in diese Signalwege eventuell ein Typus Dendritischer Zellen erzeugt werden kann, der die Vermittlung von Toleranz gegen ein bestimmtes Antigen bewerkstelligen kann.de_DE
dc.description.abstractModifications of the gene-regulating elements of the human fascin-promoter for the development of a DNA-vaccine seem to be the most promising perspective to achieve a strong DC-specific expression of the encoding gene. Duplications of certain differently sized areas which belong to 5 -distally localized homology-regions deliver a strong expression in dendritic cells while conserving the specifity for these cells. Keratinocytes which are the most prominent cell type at the application area using a gene gun for vaccination only show a weak basical expression after transfecting them transiently with the duplication constructs. Some of these constructs transfected into the murine DC cell-line XS106 lead to a similar augmentation of the expression comparable to that seen in human dendritic cells. Therefore the DNA-vaccine could also be suitable for an application in a murine model-system. In order to strengthen the nuclear transport of the plasmid repetitive consensus NFkappaB binding-sites were introduced into the vector backbone. This manipulation resulted in a strong increase in the expression level of the encoded reporter gene but was accompanied with a loss of specifity for dendritic cells. A repetitive NFkappaB binding-site taken from the human fascin-promoter which had been cloned at the same position of the plasmid had no influence on the expression level in DC, indicating that this modification is not suitable to obtain a potent DNA-vaccine. In addition, the inhibition of the NFkappaB-pathway or the MAP kinase p38 could be shown using specific pharmaceutical inhibitors in the murine DC cell-line SP37A3. This resulted in dendritic cells with a decreased expression level of the costimulatory molecules CD80 and CD86. Incubation with SB203580, a specific inhibitor of the MAP kinase p38, also showed reduced expression levels of further members of costimulatory molecules of the B7 group, CD273 and CD274. The reduced expression of these costimulatory molecules suggests a different quality of interactions with the T-cell. Intervention in these signalling pathways of the SP37A3 cells may lead to a type of dendritic cell which could be suitable for the induction of tolerance against a specific antigene.en
