dc.description.abstractIn vielen Bakterien spielt das RNA-Chaperon Hfq eine wichtige Rolle für die posttran-skriptionelle Regulation der Genexpression durch kleine nicht-kodierende RNAs. Cyanobakterielle Hfq-Homologe bilden eine besondere Unterfamilie, die zwar struktu-rell konserviert ist, aber RNA-Bindestellen mit divergierenden Eigenschaften aufweist. In Synechocystis sp. PCC6803 führt die Inaktivierung von hfq (ssr3341) zu einer Reduk-tion einer kleinen Zahl von RNAs und zu einem Defekt der Biogenese der Type IV Pili, die essentiell für die phototaktische Motilität sind. In dieser Arbeit wurden die Eigen-schaften und die subzelluläre Lokalisation von Hfq, sowie deren Auswirkungen auf die differentielle Transkriptakkumulation und das phototaktische Verhalten in diesem Modellorganismus untersucht.Eine Transkriptom-weite Microarray-Analyse zeigte, dass neben den bekannten mRNAs zwei kleine nicht-kodierende RNAs und zwei der drei CRISPR/cas-Systeme Hfq-abhängig herunterreguliert werden. Weiterhin konnte durch Mutationsstudien demonstriert werden, dass konservierte Aminosäuren auf der proximalen Oberfläche des Hfq-Hexamers, nicht aber die strukturell konservierten RNA-Bindestellen für die Funktion von Hfq essentiell sind. Während eine RNA-Bindung nicht wahrscheinlich ist, bestätigten eine Co-Immunopräzipitation und Hefe-Zwei-Hybrid-Experimente, dass Hfq mit der Sekretions-ATPase PilB1, dem Motorprotein der Typ IV Pilus Assemblie-rung, interagiert. Fluoreszenzmikroskopische Aufnahmen belegten, dass Hfq in Abhän-gigkeit von PilB1 an der Cytoplasmamembran lokalisiert ist. Gleichfalls führte die Inak-tivierung von pilB1 und pilC zu einem Verlust der Hfq-abhängigen Transkriptakkumu-lation. Demzufolge muss davon ausgegangen werden, dass die Lokalisation des Hfq-PilB1-Komplexes an die Pilusbasis essentiell für die Motilität und RNA-Akkumulation ist. Zusammengenommen deuten die Ergebnisse darauf hin, dass Hfq die Aktivität der PilB Sekretions-ATPasen in Cyanobakterien durch einen evolutionär konservierten Mechanismus auf Ebene der Protein-Protein-Interaktion reguliert.de_DE
dc.description.abstractIn many bacteria, the RNA-chaperon Hfq functions in post-transcriptional regulation of gene expression by small non-coding RNAs. Cyanobacteria harbour a subfamily of Hfq homologs that is structurally conserved but exhibits divergent RNA-binding sites. In Synechocystis sp. PCC6803, the inactivation of hfq (ssr3341) leads to a decreased accu-mulation of a small set of RNAs and abrogates the assembly of type IV pili, which are crucial for phototactic motility. In this work, the properties and the subcellular localiza-tion of Hfq as well as their implications for differential transcript accumulation and phototactic behaviour were investigated in this model organism.A transcriptome-wide microarray analysis revealed that alongside the known mRNAs two small non-coding RNAs as well as two of the three CRISPR/cas-systems are down-regulated in an Hfq-dependent manner. Furthermore, mutational analysis demonstrat-ed that several conserved amino acids on the proximal surface of the Hfq hexamer but not the structurally conserved RNA-binding sites are essential for Hfq-function. While RNA-binding seems to be unlikely co-immunoprecipitation and yeast two-hybrid ex-periments confirmed that the secretion ATPase PilB1 the motor-protein of the type IV pilus extension - is an interaction partner of Hfq. Fluorescence microscopy revealed that Hfq is localized to the cytoplasmic membrane in a PilB1-dependent manner. Con-comitantly, Hfq-dependent transcript accumulation is abrogated in a pilB1 or pilC mu-tant, indicating that localization of the Hfq-PilB1-complex to the pilus base is essential for motility and RNA accumulation. Taken together, the results suggest that Hfq might regulate the activity of PilB secretion ATPases in cyanobacteria by an evolutionary con-served mechanism based on protein-protein-interaction.en
