dc.description.abstractIn den Submerskulturen des Basidiomyceten Mycetinis scorodonius wurden die Aktivitäten von Cellulasen, Xylanasen, Laccasen, diversen Peroxidasen und Esterasen über den Kulturverlauf gemessen. Kultiviert wurde auf Stroh und den Rückständen einer Biogasanlage. Alle Enzyme zeigten ein wellenförmiges Expressionsmuster. Die Expression der Laccasen war der der beta,beta-Carotin abbauenden Peroxidasen nachgeschaltet. Bei den Kulturen mit den Rückständen einer Biogasanlage waren die Aktivitäten von Per- oxidasen und Laccasen bis zu 10 mal gegenüber den Kulturen auf Stroh erhöht. Für die ungewöhnliche beta,beta-Carotin abbauende Dyp-Typ Peroxidase MsP1 aus M. scorodonius wurde die Fähigkeit, Lignin abzubauen, mittels Messung der Partikelgrößenverteilung, Größenausschlusschromatographie, Elektronenmikroskopie und dem photometrischen Nachweis von Glucose untersucht. Für den direkten Nachweis wurde eine Suspension von organosolv Lignin eingesetzt. Als Ergebnis dieser Untersuchung stellte sich heraus, dass MsP1 in der Lage ist, sowohl ungelöste Partikel als auch gelöste Moleküle von organosolv Lignin zu modifizieren und abzubauen. Für den indirekten Nachweis wurde MsP1 als Vorbehandlungsschritt in der Verzuckerung von Stroh mittels Cellulasen und Xylanasen eingesetzt. Damit konnte eine Steigerung in der Verzuckerungsrate von Stroh erzielt werden. Ein weiteres Einsatzgebiet für MsP1 ist die Bleichung von Milchprodukten durch den Abbau von Carotinoiden, welche nach der Färbung von Käse bis zu 20 % in der Molke verbleiben. Es wurde sowohl ein zwei- und als auch ein drei-Enzym-System zur Bleichung von Molke bzw. Milch entwickelt. Durch den Einsatz von Glucoseoxidase bzw. Glucoseoxidase und Lactase konnte das für die Bleichung mittels MsP1 benötigte H2O2 in situ generiert werden. Der zeitliche Verlauf der Bleichung wurde spektralphotometrisch nach CIELab verfolgt. Des Weiteren wurde MsP1 biochemisch untersucht. Temperatur- und Druckoptimum von MsP1 wurden mittels dynamischer Differenz-kalorimetrie, Fluoreszenz- und FT-IR-Spektroskopie sowie enzymatischen Assays mit beta,beta-Carotin und 2,2´-Azino-di-(3-ethylbenzthiazolin)-6-sulfonsäure (ABTS) als Substrat untersucht. MsP1 ist thermo- und druckstabil bis zu einer Temperatur von 65 °C bzw. einem Druck von 8-10 kbar. Als Besonderheit zeigte sich eine Aktivitätserhöhung bei 500 bar, welche vermutlich auf geringe strukturelle Änderungen zurückgeht. Für einen optimalen industriellen Einsatz wurden weitere Rahmenbedingungen wie H2O2- und Lösemittel-Toleranz, pH-Optimum, sowie die kinetischen Parameter untersucht. Das pH-Optimum wurde bei pH 2,6 gefunden. MsP1 wurde schon bei geringen Konzentrationen von H2O2 gehemmt. Die inhibierende H2O2-Konzentration hängt von der Konzentration des Zweitsubstrates und des Einsatzmediums (Puffer oder Molke) ab. Die Wechselzahl (kcat) von MsP1 für die beiden Substrate beta,beta-Carotin und ABTS beträgt 0,38 ± (6,6 %) bzw. 176 ± (6,2 %) s^(-1). Zur Steigerung der Aktivität von MsP1 gegenüber beta,beta-Carotin wurden Mediatoren eingesetzt. Von den vier Mediatoren Coniferylalkohol, Ferulasäureethylester, 4-Ethylphenol und p-Coumarsäure zeigte Coniferylalkohol die höchste Steigerung der Effizienz des enzymatischen Umsatzes, dies jedoch in einem die Möglichkeit des wiederholten Einsatzes von MsP1 im industriellen Gebrauch wurde MsP1 auf Kieselgel immobilisiert.de_DE
dc.description.abstractThe present work addresses the production of cellulases, xylanases, laccases, several peroxidases, and esterases in submerged cultures of the basidiomycete Mycetinis scorodonius. The lignocelluloses from corn stover and residues from a biogas plant were used as substrates for cultivation. All enzymes showed a wave-like expression pattern. The expression of laccase followed consistently those of beta,beta-carotene-cleaving peroxidases. The activities of peroxidases and laccases were 10 times higher in cultu- res with residues from a biogas plant than in those with corn stover. The ability to degrade lignin was investigated for the beta,beta-carotene-cleaving peroxidase MsP1 from M. scorodonius. For verification, an organosolv lignin suspension was used as substrate. It was shown that MsP1 is able to degrade and modify suspended lignin particles as well as dissolved lignin molecules. For indirect evidence, corn stover was pretreated with MsP1/H2O2 followed by saccharification using cellulases and xylanases. Consequently, the yield of reducing sugars increased. Product analysis was performed by the determination of glucose using photometric assays, a determination of the particle size distribution, electron microscopy, and HPSEC analysis. A further application for MsP1 is bleaching of diary products. Carotenoids are broadly used to enhance the colour of cheese types like Cheddar and Gouda. While 80 % of the colourants are transferred into the cheese, the rest remains in the whey and impedes its commercial utilization. The ability of the fungal peroxidase MsP1 to degrade carotenoids was successfully employed for the bleaching of coloured whey and milk in fine tuned two- and three enzyme systems, respectively. The systems were composed of MsP1, glucose oxidase to generate H2O2 in situ, and if necessary, acid lactase. Time dependent bleaching was monitored colorimetrically, and the results were expressed by using the international colorimetric system of CIELab. Furthermore, biochemical properties of MsP1 were explored. Therefore, the temperature and pressure dependent stability and function of MsP1 were investigated. Several biophysical techniques such as DSC, fluorescence and FT-IR spectroscopy were combined with enzymatic studies of the enzyme with ABTS and beta,beta-carotene as substrates. The dimeric MsP1 turned out to be not only rather thermostable, but also highly resistant to pressure, i.e., up to temperatures of about 65 °C and pressures as high as 8-10 kbar at ambient temperatures. Remarkably, the activity of MsP1 increased by a factor of two until 500 kbar, which may result from slight structural changes stabilizing the transition state of the enzymatic reaction. To prepare for optimal industrial applications, basic parameters such as tolerance to H2O2 and solvents, pH-optimum, as well as steady-state kinetics were investigated. The pH-optimum was found at pH=2.6. The value of the inhibiting H2O2 concentra- tion depended on the concentration of the second substrate and the media (buffer or whey). Nevertheless, MsP1 was inhibited at low H2O2-concentrations at all conditions. Steady-state kinetics were determined for beta,beta-carotene and ABTS as substrates, kcat was determined to be 0.3 ± (6.6 %) and 176 ± (6.2 %) s^(-1), respectively. An activity enhancement of MsP1 towards beta,beta-carotene was realized by the addition of mediators. Four mediators were applied: coniferyl alcohol, ferulic acid ethylester, 4-ethylphenol, and p-coumaric acid. Coniferyl alcohol increased the catalytic efficiency to its optimum, although the effective concentration range was very narrow. Finally, MsP1 was immo- on a silica surface for the option of a reapplication of MsP1 for the industrial application.en
