dc.description.abstractSpannungsabhängige Calciumkanäle in isolierten Ganglien des Plexus myentericus neugeborener Ratten wurden pharmakologisch charakterisiert. Im Gegensatz zu früheren Arbeiten wurden isolierte Ganglien und nicht dissoziierte Neurone kultiviert, da diese nicht auf eine Rezeptorstimulation durch Neurotransmitter reagierten. Die Ganglien von drei bis elf Tage alten Ratten wurden enzymatisch isoliert und nach ca. 20 Stunden in Kultur mit dem Ca2+-sensitiven Fluoreszenzfarbstoff Fura-2-AM beladen und mit einer 35 mmol.l-1 KCl-Lösung superfundiert, um einen Anstieg der intrazellulären Ca2+-Konzentration ([Ca2+]i) via Depolarisation der Membran hervorzurufen. Dieser Anstieg der [Ca2+]i war sensitiv gegenüber Ni2+ und Co2+ ebenso wie gegenüber omega-Conotoxin MVIIA, omega-Agatoxin IVA und SNX-482. Die stärkste Hemmung wurde jedoch durch Nifedipin (5.10-7 mol.l-1) und omega-Conotoxin GVIA (4,3.10-7 mol.l-1) erreicht. Diese beiden Blocker hemmten auch den durch nicotinerge Rezeptorstimulation mittels Carbachol hervorgerufenen [Ca2+]i Anstieg. Folglich exprimieren isolierte myenterische Ganglien in Zellkultur verschiedene Typen von spannungsabhängigen Ca2+-Kanäle, von welchen der L- und der N-Typ die wichtigsten zu sein scheinen. TNFalpha Vorbehandlung dieser Ganglien über Nacht veränderte die [Ca2+]i-Antwort nach Carbachol-Gabe (5×10-5 mol×l-1), im Gegensatz zu Hyperpolarisation der Zelle und Potenzierung der durch nicotinerge Rezeptorstimulation hervorgerufenen Depolarisation, nicht signifikant. Ein direkter Einfluss von Eicosanoiden auf myenterische Neurone der Ratte konnte nur sehr bedingt gezeigt werden. Folglich scheinen Ca2+-Imaging Messungen nicht die geeignete Methode zur Untersuchung der Wirkung von Entzündungsreaktionen an enterischen Neuronen zu sein.de_DE
dc.description.abstractVoltage-dependent calcium channels of isolated ganglia from the myenteric plexus of newborn rats were pharmacologically characterised. Because they did not responde to receptor stimulation via neurotransmitters, in contrast to previous work isolated ganglia and not dissociated neurons were cultured. Ganglia of three to eleven day old rats were enzymatically isolated and loaded with the Ca2+-sensitive fluorescent dye fura-2, after about 20 hours in culture. A depolarisation-induced increase in the intracellular Ca2+ concentration ([Ca2+]i) was evoked by superfusion with 35 mmol l-1 KCl. This increase in [Ca2+]i was sensitive to Ni2+ and Co2+ as well as omega-conotoxin MVIIA, omega-agatoxin IVA and SNX-482. The strongest inhibition was achieved by nifedipine (5.10-7 mol l-1) and omega-conotoxin GVIA (4,3.10-7 mol l-1). These two blockers also inhibited the [Ca2+]i increase evoked by nicotinic receptor stimulation. Consequently, isolated myenteric ganglia in culture express different types of voltage-dependent calcium channels, from which the L- and the N-type seem to be the most important. abr>A TNFalpha pre-treatment of the ganglia over night did not alter the carbachol-induced (5×10-5 mol×l-1) [Ca2+]i response significantly, in contrast to the cellular hyperpolarisation and the potentiation of depolarisation due to nicotinic receptor stimulation. Furthermore, an only limited direct influence of eicosanoids on the [Ca2+]i of rat myenteric neurons could be shown. Consequently, Ca2+-imaging does not seem to be an appropriate method to study the action of mediators of inflammation on enteric neurons.en
