dc.description.abstractAngiogenese ist die wichtigste Voraussetzung für das Wachstum und die Metastasierungeines Tumors, da Sauerstoff und Nährstoffe für das Tumorgewebe bereitgestellt werden. DieHemmung der Angiogenese kann in der Entwicklung neuer anti-neoplastischer Therapien alseine der vielversprechendsten Forschungsmethoden angesehen werden. Zunehmend gewinnen Histondeacetylaseinhibitoren (HDIs) auf diesem Gebiet anBedeutung. Aktuelle Untersuchungen zeigen, dass HDIs antiangiogene Potenz besitzen.In diesem Zusammenhang wurde die Wirkung des neuartigen HDI alpha-Liponsäurevergleichend zu den bekannten HDIs Butyrat, Valproat und Trichostatin A in der humanenPankreaskarzinomzelllinie MIA-PaCa-2 untersucht. Die Ergebnisse zeigen, dass alpha-Liponsäure als ein wirkungsvoller HDI angesehen werden kann und in dietumorzellinduzierte Angiogenese und das Tumorwachstum von MIA-PaCa-2 Zellen eingreift. Dies zeigten zum einen Untersuchungen von Zellzahl und Proliferation von MIA-PaCa-2Zellen nach Inkubation mit alpha-Liponsäure, Butyrat, Valproat und Trichostatin A. Alle HDIshemmten dosisabhängig und weitgehend signifikant direkt die Zahl und die Proliferation derPankreaskarzinomzellen. Nach der Proliferation von Zellen kommt es zu derenDifferenzierung und zur Bildung neuer Blutgefäße. Angiogenetische Wachstumsfaktoren wieder vascular endothelial growth factor (VEGF) wurden hierbei in vielen humanenKrebszelllinien als Angiogenese-induzierend beschrieben. VEGF wird unter hypoxischenBedingungen reguliert über den hypoxia inducible factor 1 alpha (HIF-1alpha). In der vorgestelltenArbeit nahm die VEGF Sekretion dosisabhängig und meist signifikant nach Behandlung mitallen HDIs ab. Die Expression des VEGF Proteins zeigte für die HDIs alpha-Liponsäure undButyrat eine Abnahme, während die Expression des HIF-1 alpha Proteins zunahm. Anhand dieserErgebnisse kann die vermutete antiangiogene Wirkung der vorgestellten HDIs über eineHemmung des proangiogenen Faktors VEGF bestätigt werden. Diese Wirkung könnte übereine Hemmung der nukleären Translokation von HIF-1 alpha zu erklären sein, da die Ergebnisseeine verstärkte Expression des Transkriptionsfaktors im Zytoplasma nach Behandlung mitden HDIs alpha-Liponsäure und Butyrat zeigten. In Anlehnung an diese Ergebnisse kann die Hypothese aufgestellt werden, dass HDIs HIF-1 alphabeeinflussen und somit auch den für die Angiogenese bedeutenden Faktor VEGF, derwiederum über HIF-1 alpha reguliert wird. Weiterhin wurde der Einfluss des Vitamin D Rezeptors (VDR) in MIA-PaCa-2 Zellenuntersucht. VDR hemmt die Angiogenese in vitro und in vivo. Nach Behandlung mit den HDIs alpha-Liponsäure und Butyrat kam es zu einer gesteigerten Expression von VDR und somitsynergistischen Verstärkung der Wirkung in MIA-PaCa-2. Um die Rolle der mitogenactivatedprotein kinases (MAPKs) in der HDI-gesteuerten Zelldifferenzierung und VDR Expressionzu beurteilen, wurde der spezifische MAPK-Inhibitor SB203580 verwendet.Die vorliegenden Ergebnisse machen deutlich, dass die verwendeten HDAC-Inhibitoren zueiner Phosphorylierung der p38 MAPK und dabei möglicherweise einer Zunahme der VDRExpression und Zelldifferenzierung führen, wohingegen eine Hemmung der p38 MAPK diep38-Phosphorylierung reduziert. Zum Schluss konnte gezeigt werden, dass der p38 MAPK Signalweg bei denPankreaskarzinomzellen in die Kontrolle der VEGF Synthese involviert ist. Die Ergebnissebestätigen eine Hemmung der VEGF Proteinexpression in MIA-PaCa-2 nach Behandlung mit& #945;-Liponsäure und Butyrat während diese Wirkung unter Verwendung des spezifischen p38MAPK Inhibitors SB203580 nicht zu beobachten war.de_DE
dc.description.abstractAngiogenesis is essential for tumour growth and metastasis, because it provides oxygen andnutrients to the tumour tissue. Inhibition of angiogenesis is considered to be one of the mostpromising strategies in the development of novel anti-neoplastic therapies. Histondeacetylaseinhibitors (HDIs) has gained importance in recent studies and also hasbeen shown to be a potent antiangiogenic agent. Recent examinations show that HDIs arepotent antiangiogenic agents. In this context the effects of alpha-lipoic acid, a novel inhibitor of histondeacetylases, comparedwith the well-known HDIs (butyrate, valproate and trichostatin A) in human pancreaticcarcinoma cells (MIA-PaCa-2) is studied. The results indicate that alpha-lipoic acid is an effectiveHDI and impairs the tumour cell induced angiogenesis and tumour growth of MIA-PaCa-2cells. The above finding was shown first in examinations of cell count and proliferation of MIAPaCa-2 cells after incubation with alpha-lipoic acid, butyrate, valproate and trichostatin A. AllHDIs inhibited directly cell count and proliferation of pancreatic carcinoma cells, mostsignificantly in a dose-dependent manner. After their proliferation, tumour cells differentiate toform a new vessel. Angiogenic growth factors such as the vascular endothelial growth factor(VEGF) have been observed in many human cancer cell lines inducing angiogenesis.Regulated by hypoxia the VEGF gene is under the control of the hypoxia inducible factor 1 alpha(HIF-1 alpha). In the present work a dose-dependent and most significantly decreased secretionof VEGF after treatment with HDIs is shown. The expression of the VEGF protein wasreduced while that of the HIF-1 alpha protein expression increased after HDI treatment. On basisof these results we conclude, that the antiangiogenic effects of the tested HDIs are causedby the inhibition of the proangiogenic factor VEGF. This effect could be explained byinhibiting the HIF-1 alpha nuclear translocation, as the results showed enhanced cytoplasmicexpression of the transcription factor after incubation with alpha-lipoic acid and butyrate.Summarizing these findings hypothesize, that HDIs influence HIF-1 alpha and subsequently theangiogenesis related factor VEGF, regulated by HIF-1 alpha. Furthermore the influence of the vitamin D receptor (VDR) in MIA-PaCa-2 cells is studied.VDR inhibits angiogenesis both in vitro and in vivo. After treatment with HDIs, alpha-lipoic acidand butyrate, an upregulation of VDR and synergistic amplification of the effect in MIA-PaCa-2 could be observed. To assess the role of mitogen-activated protein kinases (MAPKs) inHDI-mediated cell differentiation and VDR-expression the specific p38 MAPK inhibitorSB203580 was used. The present study revealed that the tested HDIs cause phosphorylation of p38 MAPK andthereby possibly upregulation of VDR expression and cell differentiation, whereas inhibitionof p38 MAPK abolishes the p38-phosphorylation. In conclusion, we demonstrated that p38 MAPK pathway is involved in the synthesis ofVEGF in pancreatic carcinoma cells. The findings also confirm the inhibition of VEGF proteinexpression in MIA-PaCa-2 after treatment with α-lipoic acid and butyrate while these effectswere not observed using the specific p38 MAPK inhibitor SB203580.en
