dc.description.abstractÜber die Expression des hypophysären POMC-Gens und die Freisetzung von POMC-Derivaten aus der Hypophyse liegen sehr viele Informationen vor. 'Streß' im Sinne physischer und psychischer Belastung spielt für die Freisetzung der hypophysären POMC-Derivate eine große Rolle. Auch zur Expression des POMC-Gens und zur Freisetzung von POMC-Derivaten in der Haut gibt es eine beträchtliche Anzahl von Untersuchungen. Die Funktion der hypophysären wie der dermalen POMC-Derivate ist jedoch weitgehend unklar, auch wenn eine Reihe von Effekten von ACTH, alpha-MSH oder ß-Endorphin bekannt sind. Die Expression des hypophysären POMC in der Haut und die Freisetzung von hypophysären POMC-Derivaten (obligatorisch aus dem Exon 3-Bereich) sind 'anerkannte Hypothese' auf dem betreffenden Forschungsgebiet. Neuerdings wird diese 'Lehrmeinung' jedoch angegriffen und dieser die Hypothese eines (bezogen auf das hypophysäre) aberranten POMC-Gens bzw. eines aberranten POMC-Proteins in Keratinozyten und Melanozyten gegenübergestellt. In dieser Arbeit konnte an der Keratinozytenzellinie HaCaT weder die Expression von hypophysärem POMC noch die Bildung oder Freisetzung hypophysärer POMC-Derivate nachgewiesen werden. An der Epidermoidzellinie A 431 konnte die Expression des hypophysärem POMC ebenfalls nicht nachgewiesen werden. Lediglich der Einsatz eines der Primerpaare aus dem Bereich des Exon 3 ergab ein sehr schwach positives Signal; diese Befunde sind, insgesamt betrachtet, mit der Existenz einer Variante des hypophysären POMC-Gens in A 431-Zellen, wie sie in einer früher von der Arbeitsgruppe um Farooqi publizierten Arbeit bereits vorgeschlagen wurde (Can et al., 1998), kompatibel. Auch die Bildung und Freisetzung der hypophysärer POMC-Derivate konnte bei A 431-Zellen nicht nachgewiesen werden, was wiederum mit der Existenz eines 'aberranten' POMC-Gens vereinbar ist. Die beiden für den Nachweis von alpha-MSH-IRM bzw. CLIP/ACTH-IRM entwickelten RIAs erfüllten ihre Aufgabe zuverlässig und stehen für weitere Versuche zur Verfügung. Unsere Befunde stützen somit bis jetzt nur die von der Gruppe um Farooqui aufgestellte Hypothese: Sowohl der häufig erhobene Befund eines human-hypophysären POMC-Gens in der Haut als auch unsere diesbezüglich negativen Befunde an Zellinien sind kompatibel mit der Expression eines -bezogen auf das hypophysäre POMC-Gen- aberranten POMC-Gens in der Haut.de_DE
dc.description.abstractPlenty of information is available on the expression of pituitary POMC gene and the release of POMC-derived peptides from the pituitary. Physiological and psychological 'stress' shows signficant impact on the release of POMC-derived peptides. Like wise, plenty of research has been conducted on the expression of the POMC gene and on the release of POMC-derived peptides in the skin. However, despite the number of proven effects of ACTH, alpha-MSH and ß endorphin, the function of pituitary and cutaneous POMC-derived peptides is largely unclear. In the relevant field of POMC-research the expression of pituitary POMC in the skin and the release of POMC-derived peptides (always from the exon 3 region of POMC gene) constitute 'common and accepted hypothesis'. They are, however, recently challenged when contrasting them with a divergent hypothesis about an isoform or variant of the (with regard to the pituitary) POMC gene or, respectivly, a variant of the POMC-protein in keratinocytes and melanocytes. Our research could not confirm either the expression of pituitary POMC or the production and release of pituitary POMC-derived petides at the keratinocyte cell line HaCaT. The expression of pituitary POMC could also not be confirmed at the epidermal carcinoma cell line A 431. A weak positive signal could solely be produced by using primers of the exon 3 region of human POMC. Our research results prove in total compatible with the previously published opinion by researcher group around Farooqui (Can et al., 1998) about the existence of an isoform or variant of the pituitary POMC gene in A 431-cells. The fact that we could also not confirm the production and release of pituitary POMC peptides in A 431-cells is also regarded coherent with the existence of an isoform or variant of the POMC gene. Both radioimmunoassays developed to test and confirm the existence of alpha-MSH-IRM, respectivly CLIP-IRM, functioned properly, and can be utilized for further research projects. Thus, our findings support so far only the hypothesis put forth by the researcher group around Farooqui: Both, the often stated existence of a human-pituitary POMC gene in the skin and our negative research results on the cell lines are coherent with the expression of a (with regard to the pituitary POMC gene) variant of the POMC gene in the skin.en
