dc.description.abstractIn der vorliegenden Arbeit konnte erstmals gezeigt werden, dass Sphingosin-1-Phosphat (S1P) in NIH3T3- und C3H10T1/2-Mäusefibroblasten einen spannungsunabhängigen, calciumabhängigen Kaliumstrom transient aktiviert, der durch Charybdotoxin, Margatoxin und Iberiotoxin vollständig blockiert werden kann. Die Kaliumstromaktivierung führt zu einer starken Hyperpolarisation der Zellen. Der S1P-aktivierte Kaliumstrom hat identische elektrophysiologische und pharmakologische Eigenschaften wie der durch Lysophosphatidsäure (LPA) aktivierte Kaliumstrom in NIH3T3-Zellen. In der Whole-Cell Ableitung führt nur die erste Applikation von S1P zu einer transienten Kaliumstromaktivierung, wohingegen in der Cell-Attached Ableitung mehrfache transiente Kaliumstromaktivierungen möglich sind. Der transiente Stromverlauf und das Ausbleiben einer zweiten Kaliumstromaktivierung im Whole-Cell Modus sind nicht auf eine Inaktivierung der Kaliumkanäle zurückzuführen. Jeweils identische, transiente Kaliumstromaktivierungen treten auf, wenn S1P und LPA bzw. LPA und S1P nacheinander appliziert werden. Dieser Befund weist darauf hin, dass S1P und LPA bezüglich der Kaliumstromaktivierung keine Cross-Desensitisierung zeigen. In C3H10T1/2-Mäusefibroblasten, die die nicht-rezeptorgekoppelte Proteintyrosinkinase c-Src überexprimieren, ist die Amplitude des S1P-induzierten Kaliumstroms fast doppelt so hoch wie in den Kontrollzellen. Die Expression einer nicht-myristilierbaren c-Src Mutante führt zu einer weiteren Erhöhung der Kaliumstromantwort, während die Expression einer Kinase-defekten c-Src Mutante die Stromantwort auf 43 % des Kontrollwertes reduziert. Diese Daten zeigen, dass S1P in Mäusefibroblasten calciumabhängige Kaliumkanäle über eine intrazelluläre Signalkaskade aktiviert, in die c-Src eingeschaltet ist. Weiterhin konnte gezeigt werden, dass c-Src auch für die Kaliumkanalaktivierung durch LPA eine entscheidende Rolle spielt. In der vorliegenden Arbeit wurde erstmals nachgewiesen, dass LPA in glatten Gefäßmuskelzellen (A7r5) einen L-Typ Calciumstrom inhibiert. Pertussistoxin (PTX) vermindert signifikant die LPA-induzierte Blockade dieses Stroms, was auf die Beteiligung eines PTX-sensitiven G-Proteins schließen lässt. Das für L-Typ Calciumströme typische elektrophysiologische Profil wurde nur an solchen A7r5-Zellen gefunden,die vollkommen isoliert lagen und keinen Membrankontakt zu Nachbarzellen hatten. In weiteren Experimenten muss nun untersucht werden, in welche biologischen Prozesse die S1P- und LPA-modulierten Ionenkanäle eingebunden sind.de_DE
dc.description.abstractThe present study shows for the first time that in NIH3T3 and C3H10T1/2 mouse fibroblasts sphingosine-1-phosphate (S1P) transiently activates a voltage-independent, calcium-dependent potassium current that can be completely blocked by charybdotoxin, margatoxin, and iberiotoxin. The potassium current activation leads to a large membrane hyperpolarization. The S1P-activated potassium current has identical electrophysiological and pharmacological properties compared to the lysophosphatidic acid (LPA)-activated potassium current in NIH3T3 cells. In the whole-cell recording mode, only the first application of S1P leads to a transient potassium current activation, whereas in the cell-attached recording mode several transient potassium current activations are possible. The transient current response and the failure of S1P to evoke a second potassium current response in the whole-cell recording mode are not due to an inactivation of potassium channels. Identical transient potassium current activations are evoked when S1P and LPA or LPA and S1P are applied consecutively. These findings indicate that in the case of potassium current activation no cross-desensitization occurs by S1P and LPA. In C3H10T1/2 mouse fibroblasts that overexpress the non-receptor protein tyrosine kinase c-Src, the amplitude of the S1P-induced potassium current is almost doubled compared to the one found in control cells. Expression of a non-myristylated c-Src mutant leads to a further increase in the potassium current response, whereas expression of a kinase-defective c-Src mutant reduces it to 43 % compared to the control value. These data show that S1P activates calcium-dependent potassium channels in mouse fibroblasts via an intracellular signalling pathway that involves c-Src. Furthermore, it was demonstrated that c-Src plays also an crucial role for the potassium channel activation by LPA. A further finding of the present study is that in vascular smooth muscle cells (A7r5) LPA inhibits an L-type calcium current. Pertussis toxin (PTX) significantly reduces the LPA-induced block of this current, which indicates the involvement of a PTX-sensitive G protein. The typical electrophysiological profile of L-type calcium channels was only found in A7r5 cells lying completely isolated without any contacts to neighbouringcells. In further experiments it should be investigated in which biological activities the S1P and LPA-modulated ion channels are involved.en
