dc.description.abstractZiele: Die Ca2+-Konzentrationserhöhung ist ein Kofaktor von vielen Zellprozesseneinschließlich Apoptose und Nekrose. Die Hypothese von dieser Studie war, dass blutigerzerebrospinaler Liquor (bCSF) von Patienten nach intraventrikulärer Blutung einen Ca2+-Anstieg verursachen konnte, der Apoptose oder Nekrose in humanen Astrozyteninduziert.Methode: Humane Astrozyten wurden mit bCSF inkubiert. In Kontrollexperimenten wurdenativer zerebrospinaler Liquor (nCSF) verwendet.Die zytosolischen Ca2+-Konzentration von den einzelnen Zellen wurde mit dem Ca2+-sensitiven Farbstoffe (Fura-2) gemessen. Vier Blocker wurden verwendet: Nimodipin (LTyp-Calciumkanale Hemmer), 2-aminoethoxydiphenyl-borate (2APB) (IP3-Rezeptorenblocker des endoplasmatischen Retikulums), Suramin (P2-Rezeptorenantagonist), D (-)-2-amino-5-phosphonopentanoic-acid (2A5P) (NMDARezeptorblocker),und Cyclosporin A (MPTP-Inhibitor). Zur Messung von Nekrose undApoptose wurden Astrozyten mit Hoechst-33342 und Propidium Iodid markiert.Die Ergenisse der vorgelegten Arbeit zeigen:1) dass blutiger zerebrospinaler Liquor Nekrose in humanen Astrozyten induziert unddass die zytosolische Ca2+-Konzentrationserhöhung eine wichtige Rolle bei derEntstehung von bCSF-induzierter Nekrose spielt,2) dass die zytosolische Ca2+-Konzentrationserhöhung und die Nekrose von derAktivierung P2-Rezeptoren und IP3-abhängiger Ca2+-Kanäle amendoplasmatischen Retikulum abhängig sind,3) dass eine Blockade der P2-Rezeptoren durch Suramin den Ca2+-Anstieg und dieNekroserate reduziert, während NMDA-Blockade durch D(-)-2-Amino-5-Phosphonopentanoic Acid keine signifikante Veränderung auf den Ca2+-Anstiegbzw. die Nekroserate bewirkt,4) dass der ATP-Gehalt in bCSF gegenüber nCSF um ungefähr das 400-facheerhöht ist,5) dass eine Hemmung der Ca2+-Kanäle vom L-Typ durch Nimodipine zu einergeringen aber gerade signifikanten Verringerung des Ca2+-Anstiegs und derNekroserate führt,6) dass eine Blockade der IP3-Rezeptoren durch 2APB den Ca2+-Anstieg und dieNekroserate reduziert, und dass eine Blockade der MPTP durch Cyclosporin A zursignifikanten Nekrose-Reduktion ohne Einfluss auf die zytosolische Ca2+-Kinetikführt.Schlussfolgerung:Blutiger zerebrospinaler Liquor verursacht Nekrose in humanen Astrozyten. Die bCSFinduzierteNekrose wird durch einen zytosolischen Ca2+-Anstieg verursacht, der durchCa2+-Freisetzung aus dem ER zustande kommt. dc.description.abstractAims: An increase in cellular Ca2+ concentration is a cofactor of many cellular processesincluding apoptosis and necrosis. This studyۥ s hypothesis was that hemorrhagiccerebrospinal fluid (CSF) from patients after intraventricular hemorrhage may cause anincrease in Ca2+ concentration which induces apoptosis or necrosis in an in-vitro model ofhuman cerebral astrocytes.Method: Human astrocytes were incubated with CSF from patients with intraventicularhemorrhage. In control experiments native CSF was used.Cytosolic Ca2+-concentrations were measured in single cells by fura-2. Fourpharmakological blockers were used: Nimodipine, 2-aminoethoxydiphenyl borate (2APB),suramine, D (-)-2-Amino-5-Phosphonopentanoic-Acid (2A5P), and cyclosporin A, thatblock the L-type Ca2+-channel, the endoplasmic reticulum Ca2+ channels, the ATPsensitivePY2-receptor, the NMDA-receptor, and the mitochondrial permeability transitionpore (MPTP), respectively. Apopotosis and necrosis were evaluated by staining the cellswith Hoechst-3342 and propidium iodide.The major findings of this study are :1) the fact that bloody cerebrospinal fluid induces necrosis and not apoptosis inhuman astrocytes. The increase of cytosolic Ca2+-concentration plays an importantrole in the bCSF-induced necrosis.2) the fact that the increase of cytosolic Ca2+-concentration and the necrosis aredependent on the activation P2 receptors and IP-3 dependent Ca2+ channels ofthe endoplasmic reticulum.3) the fact that blocking the P2 receptors by suramin reduces the Ca2+ increase andthe necrosis rate, while blocking of NMDA receptors by D (-)-2-Amino-5-phosphonopentanoic acid causes no significant change in the Ca2+ increase or thenecrosis rate.4) the fact that the ATP content is 400-fold higher in bCSF as compared to nCSF.5) the fact that an inhibition of the L- type Ca2+ channels with Nimodipine leads to asignificant reduction of the Ca2+ increase and the rate of necrosis.6) the fact that a blockade of the IP3 receptors by 2APB reduces the Ca2+ increaseand the necrosis rate, and that a blockade of the MPTP by Cyclosporin A leads tosignificant reduction of the necrosis without significant influence on cytosolic Ca2+kinetics.Conclusion:Bloody CSF induces necrosis but not apoptosis in human astrocytes. The CSF-inducednecrosis is due to increase of cytosolic Ca2+ which is mediated through capacitative Ca2+release by the ER involving the ATP-sensitive PY2-receptor.en
