dc.description.abstract | Diese Arbeit beschäftigt sich mit dem Hepatitis C Virus (HCV), welches spezifisch die Leber befällt und bedeutsamster Auslöser für die chronische Hepatitis ist. Es wurde gezeigt, dass Nukleotid-Insertionen innerhalb der HCV-5´-UTR die microRNA-(miR)-122-vermittelte Stimulierung der Translation reduzieren. Zudem wurde untersucht, ob mittels Sucrose-Dichtegradienten-Ultrazentrifugation ein Einfluss der miR-122 auf die Bildung der Translations-Initiationskomplexe nachgewiesen werden kann. Das Hepatitis C Virus (HCV) wird in die Familie der Flaviviridae klassifiziert und ist Auslöser der NonA-NonB-Hepatitis. Als einzelsträngiges RNA-Virus mit Positiv-Orientierung kann es intrazellulär direkt translatiert werden, gleichwohl es nicht capabhängig, sondern vermittelt über die HCV-5´-IRES translatiert wird. Das Genom enthält einen offenen Leserahmen (ORF) und kodiert für ein einziges Polyprotein, welches co- und posttranslational in Struktur- und Nicht-Struktur-Proteine prozessiert wird. Der ORF wird flankiert von 5´-UTR und 3´-UTR. Die 5´-UTR ist unter den HCVIsolaten hochkonserviert und beinhaltet unter anderem zwischen den Stem-Loops I und II die beiden miR-122-Bindestellen. Die leberspezifische miR-122 stimuliert die HCV-Translation. Das Einfügen von Nukleotiden zwischen zweiter miR-122-Bindestelle und Stem-Loop II der HCV-5´-UTR zeigt eine signifikante Reduktion der Stimulierung der Translation durch miR-122 in HuH-7-Zellen, HuH-7.5-Zellen sowie unter Verwendung von d(T)-miR-122. Die Stimulierung bleibt in sämtlichen Versuchsreihen bei einer Insertion von 20 Nukleotiden komplett aus. Dieses Ergebnis zeigt, dass der Abstand zwischen miR-122-Bindestellen und der HCV-IRES für die Funktionalität der Stimulation der HCVTranslation durch miR-122 wichtig ist. Zudem wurde in dieser Arbeit die Sucrose-Dichtegradienten-Ultrazentrifugation für HuH-7-Zellen optimiert und mit dieser Methode der Einfluss der miR-122 auf die Bildung der Translations-Initiationskomplexe untersucht. dc.description.abstract | This study is about Hepatits C Virus (HCV), which specifically infects the liver and which is an important causative agent of chronical hepatitis. It is shown that nucleotide insertions in the 5´-UTR can reduce stimulation of HCV translation by microRNA-(miR)-122. Additionally, the formation of HCV initiation complexes during translation was investigated by analysis of sucrose density gradients after ultracentrifugation. The Hepatitis C Virus is classified into the family of Flaviviridae and is the main causative agent of NonA-NonB hepatitis. Its single-stranded RNA virus genome can be directly translated in the cytoplasm because of its positive orientation. The translation is mediated cap-independently by the 5´-IRES which allows a direct interaction of the ribosome with the HCV RNA. The RNA genome consists of the open reading frame (ORF) and codes for one single polyprotein that is co- and posttranslationally processed into structure and non-structure proteins. The ORF is flanked by the 5´-UTR and the 3´-UTR. Among all HCV isolates the 5´-UTR is highly conserved and contains two possible miR-122 target sites. The liver-specific miR-122 stimulates HCV translation. By increasing the distance between the second miR-122 target sequence and the base of the stem-loop II through insertion of nucleotides the miR-122 stimulation of HCV translation can be reduced in HuH-7-cells and HuH-7.5-cells, even under application of (dT)-miR-122. The insertion of 20 nucleotides completely abolishes miR-122 stimulation of HCV translation. These results show that the distance between the miR-122 binding sites and the HCV-IRES is important for the stimulation of HCV translation by the miR-122. Moreover, the ultracentrifugation of sucrose density gradients was optimized for HuH-7-cells and is used to analyze the formation of initiation complexes during HCV translation. It is shown that miR-122 does not have any influence on steady-state level of complex formation during HCV translation initiation. | en |
