dc.description.abstractZahlreiche Arbeiten zeigen, dass die MPTP eine wichtige Rolle für die Entstehung des myokardialen Reperfusionsschadens spielt. In unserer Arbeitsgruppe wurde bereits gezeigt, dass Ca2+-Oszillationen aus dem sarkoplasmatischen Retikulum (SR) eine wichtige Rolle für die reperfusionsinduzierte Hyperkontraktur und Nekrose der Herzmuskelzellen spielt. Das Ziel der vorliegenden Arbeit ist zu klären, ob und wann eine MPTP-Öffnung während der Ischämie und Reperfusion stattfindet, welche Rolle die sarkoplasmatisch abhängigen Ca2+-Oszillationen und die MPTP-Öffnung bei der Entwicklung der Hyperkontraktur spielen, und ob der Verlust der mitochondrialen Ca2+-Sequestrierung bei der MPTP-Öffnung die SR-abhängigen Ca2+-Oszillationen verstärkt.Unsere Untersuchungen haben wir an isolierten Herzmuskelzellen adulter Wistar Ratten in vitro durchgeführt. Die Zellen wurden mit Fluoreszenz-Indikatoren vorinkubiert, um die zytosolische und mitochondriale Calcium-Homoöstase sowie die MPTP-Öffnung zu identifizieren. Zur Bestimmung der Ca2+-Homoöstase wurden die Zellen mit Fura-2 AM beladen. Zur Untersuchung der MPTP-Öffnung haben wir die Zellen mit Calcein-AM oder mit JC-1 beladen. Die Kardiomyozyten wurden mit einer physiologischen Lösung perfundiert, dann wurden sie 90 Minuten lang einer ischämischen Phase (Anoxie) unterzogen, die von einer Reperfusionsphase (Reoxygenation) gefolgt wurde. In der Reperfusionsphase wurden Inhibitoren der MPTP (Cyclosporin A) oder SR-Ca2+-Freisetzung (Ryanodin) oder mitochondriale Ca2+-Aufnahme (Ru 360) appliziert.Unter Kontrollbedingungen kommt es zu einem irreversiblen Verlust der mitochondrialen Calceinkonzentration und einem irreversiblen Kollaps des mitochondrialen Membranpotentials. Die Behandlung der Kardiomyozyten mit Cyclosporin A , Ru 360 oder Ryanodin in der Reperfusionsphase führt zu signigikant geringerem Calceinverlust und verhindert den Kollaps des mitochondrialen Membranpotentials. Die mitochondriale Calciumkonzentration steigt in der Ischämie an. Mit Beginn der Reperfusion kommt es zu einem irreversiblen Anstieg. Die Behandlung mit Cyclosporin A, Ru 360 oder Ryanodin in der Reperfusionsphase führt zur Erholung der mitochondrialen Calciumkonzentration. Die Zellen entwickeln in der Reperfusionsphase unter Kontrollbedingungen Ca2+-Oszillationen und Hyperkontraktur, die durch die Behandlung mit Ryanodin, Ru 360 oder Cyclosporin A signifikant verringert wurden.Die Ergebnisse der Arbeit zeigen, dass die MPTP-Öffnung in der frühen Phase der Reperfusion, d.h. bereits in den ersten Minuten stattfindet. Die MPTPÖffnung ist auf eine mitochondriale Ca2+-Überladung zurückzuführen, die durch zytosolische Ca2+-Oszillationen ausgelöst wird, welche wiederum durch eine primäre Ca2+-Aufnahme ins sarkoplasmatische Retikulum verursacht werden. Die MPTPÖffnung führt durch Interaktion mit dem sarkoplasmatischen Retikulum zu einer Verstärkung der zytosolischen Ca2+-Oszillationen.de_DE
dc.description.abstractSeveral studies showed that the mitochondrial permeability transition pores play an important role in the development of a myocardial reperfusion injury. We have already shown that Ca2+-oscillation depending on the sarcoplasmic reticulum (SR) plays an important role in the development of the hypercontraction and nekrosis of the cardiomyocytes. The aim of the present study is to investigate whether and when do mitochondrial permeability transition pores opening occur during ischemia and reperfusion. The focus of the study was the role of the Ca2+-oscillations in the development of the hypercontracture and MPTP opening and if a loss of mitochondrial Ca2+ through MPTP opening reinforce the SR dependent Ca2+-oscillations.The experiments were done in isolated cardiomyocytes of adult 200-300 g Wistar rats in vitro. To determine the cytosolic or mitochondrial Ca2+-concentration, we loaded the cells with Fura-2 AM. To investigate the opening of MPTP we loaded the cells with Calcein-AM or JC-1. The cardiomyocytes were perfused with physiological solution and then they were subjected to 90 minutes ischemic phase (anoxia) which was followed by reperfusion phase (reoxygenation).The inhibitors of MPTP (Cyclosporin A) or SR-Ca2+-release (Ryanodin) or mitochonndrial Ca2+ uptake (Ru 360) were applied during reperfusion Under control conditions the cardiomyocytes show an irreversible loss of mitochondrial calcein concentration and an irreversible collapse of mitochondrial membrane potential. Treatment of cardiomyocytes with cyclosporine A, Ru 360 or ryanodine during reperfusion prevents mitochondrial calcein loss and improves recovery of mitochondrial membrane potential. During ischemia, mitochondrial calcium concentration rises. In early reperfusion a further irreversible increase in mitochondrial calcium occurs. Treatment with cyclosporine A, Ru 360 or ryanodine during reperfusion leads to a complete recovery of mitochondrial calcium concentration. In early reperfusion cardiomyocytes develop Ca2+-oscillations and hypercontracture. These were significantly reduced by treatment with ryanodine, Ru 360 or cyclosporin A.The major findings of the study are that mitochondrial permeability transition pores open in the early reperfusion phase, i.e. within the first minutes of the reperfusion. The MPTP opening occurs due to a mitochondrial Ca2+-overload that is triggerd by the Ca2+-oscillations originating from the sarcoplasmic reticulum. The MPTP opening is a part of an interaction between sarcoplamic reticulum and mitochondria reinforcing the cytosolic Ca2+-oscillations.en
