dc.description.abstractIn der Dissertationsarbeit von Xiao (Xiao, 2003) ergaben sich Hinweise darauf, dass das Sekretoglobin SCGB 2A1, ein typisches Sekretprotein der Prostata, sowohl in den Basalzellen wie auch den Luminalzellen der humanen Prostata exprimiert wird. Bislang wurden jedoch die sezernierten Proteine der Prostata ausschließlich in den Luminalzellen gefunden. Darüber hinaus fand Xiao SCGB 2A1 sowohl im Epithel der fetalen und adulten Prostata wie auch im Prostatakarzinom, jedoch konnte das Protein in einer benignen Prostatahyperplasie nicht nachweisen werden. Möglicherweise könnte SCGB2A1 daher einen Marker darstellen, mit dem es möglich ist Karzinom von BPH zu unterscheiden. Ziel dieser Arbeit war es daher, die Expression in den Basalzellen mit Hilfe der doppelimmunfluoreszenzmikroskopischen Analyse abzuklären und die Expression in BPH Proben mittels Immunhistochemie zu überprüfen.Die durchgeführten Untersuchungen ergaben, dass SCGB 2A1 gleichermaßen in den Basalzellen wie den Luminalzellen der Prostata vorliegt. Damit scheint es das erste sezernierte Prostataprotein zu sein, das in beiden Zelltypen nachzuweisen ist. Dieser Befund unterstützt andere Berichte, die Ähnlichkeiten im Expressionsprofil von Basal- und Luminalzellen gefunden haben, wie auch die These, dass beide Zelltypen sich aus einer gemeinsamen Stammzelle entwickeln.Darüber hinaus wurde SCGB 2A1 im Drüsenepithel sowohl von BPH wie auch Prostatakarzinomproben nachgewiesen. Das Protein stellt damit also keinen spezifischen Karzinommarker dar. Es ergaben sich jedoch Hinweise darauf, dass SCGB 2A1 bei Prostatakarzinom wie auch PBH im Vergleich zu gesundem Gewebe überexprimiert wird. Dies wurde bereits für ein anderes Sekretoglobin, das Mammaglobin (SCGB 2A2), beim Mammakarzinom vielfach gezeigt, so dass es heute als etablierter Tumormarker gilt. Es könnte sich also lohnen, auch die Überexpression von SCGB 2A1 bei BPH und Prostatakarzinom genauer und mit anderen geeigneten Methoden wie der qPCR zu untersuchen, um zu überprüfen, ob das Protein nicht als Tumormarker diagnostischen Wert hat.de_DE
dc.description.abstractThe Secretoglobin SCGB 2A1, a typical secretory protein of the prostate, can be detected in the basal as well as luminal cells of the human prostate. To date, secreted proteins were only found to be expressed by the luminal cells. Moreover, Xiao (2003) found SCGB 2A1 in the epithelium of the fetal and adult prostate as well as in prostate carcinoma, but could not detect the protein in a sample of benign prostate hyperplasia (BPH). Therefore, SCGB 2A1 could represent a valuable diagnostic marker protein that is able to distinguish between prostate carcinoma and BPH. Aim of this thesis was therefore to verify expression of SCGB 2A1 in basal cells by using double immunofluorescence and to survey expression in BPH samples by immunohistochemistry.The results show that SCGB 2A1 is present in basal and prostate cells alike. This identifies SCGB 2A1 as the first secreted prostate protein that can be demonstrated in both cell types. This finding supports other publications reporting similarities in the expression profiles of basal and luminal cells and the hypothesis that both cell types develop from a common precursor stem cell.Moreover, SCGB 2A1 could be detected in the glandular epithelium of BPH as well as in prostate carcinoma samples. Therefore, it does not qualify as carcinoma-specific marker. However it appears that SCGB 2A1 is overexpressed in prostate carcinoma and BPH samples in comparison to normal tissue. This was also shown for another secretoglobin mammaglobin (SCGB 2A2) that is specifically overexpressed and secreted into the circulation by breast cancer cells and represents an established tumor marker for this malignancy. Therefore, it would be worthwhile to thoroughly investigate overexpression of SCGB 2A1 in BPH and prostate cancer with adequate methods like qPCR in order to find out if the protein may qualify as a diagnostic marker for prostate cancer.en
