dc.description.abstractDas Glioblastom (WHO Grad IV) ist der häufigste maligne Hirntumor des Erwachsenen und geht trotz multimodaler Therapie mit einer mittleren Überlebenszeit von nur 12-15 Monaten einher. Das Glioblastom zeichnet sich durch eine ausgeprägte intratumorale morphologische, phänotypische und zelluläre Heterogenität u.a. mit Krebsstammzellen aus, die für die Tumorentstehung, Rezidivbildung und Therapieresistenz verantwortlich gemacht werden. Krebsstammzellen befinden sich in spezifischen perivaskulären und hypoxischen Nischen. Auf der Suche nach neuen Therapieansätzen hat man in letzter Zeit den Fokus auf microRNA´s gerichtet, die essentielle pro-tumorigene Eigenschaften wie Angiogenese, Zelldifferenzierung und -migration fördern und die zelluläre Adaptation an Hypoxie begünstigen.Ziel dieser Arbeit war es, neue Schlüsselmechanismen zu identifizieren, die hypoxieinduziert die Tumormalignität im Glioblastom fördern. Hierbei konnten wir eine wichtige pro-tumorigene Rolle der Hypoxie/HIF-induzierten miR210 im Glioblastom identifizieren. Methodisch wurde die miR210 Expression über lentivirale Gain - und loss-of-function Experimente in vitro und in vivo in humanen primären und etablierten Glioblastomzellen unter normoxischen (21% O2) und hypoxischen (1% O2) Bedingungen moduliert. Die Überexpression von miR210 förderte die Stammzelleigenschaften in Glioblastomen und erhöhte die Expression des Tumorstammzellmarkers CD133 auf Protein- bzw. mRNA- Ebene. Gleichzeitig aktivierte miR210 eine erhöhte Expression von HIF-1a, HIF-2a und HIF Targetgenen (VEGF, Carboanhydrase IX). In Übereinstimmung mit einer pro-tumorigenen Rolle führte die Überexpression von miR210 zur einer signifikanten Zunahme des Tumorvolumens und der Gefäßdichte in zwei intrakraniellen Xenograftmodellen der Maus (GMB046X, G55 GBM). Hingegen reduzierte die Inhibition von miR210 über lentivirale Überexpression von antisense Oligonukleotiden den Tumorstammzellphänotyp und das Tumorwachstum.Schlüsselerkenntnis der vorliegenden Arbeit ist, dass miR210 eine wichtige pro- tumorigene Rolle im Glioblastom zukommt und dass die HIF-1 vermittelte miR210 Expression als Teil eines feedforward loops HIF abhängige Tumorcharakteristika wie Angiogenese oder den Tumorstammzellphänotyp, die essentiell für die Tumorprogression sind, vorantreibt. Unsere Befunde legen nahe, dass miR210 als potentieller Therapieansatzpunkt zu einer gezielten Eliminierung von Gliomstammzellen in hypoxischen Nischen sowie zur Inaktivierung des pro-tumorigenen HIF-Signalwegs herangezogen werden kann.de_DE
dc.description.abstractGlioblastoma (WHO grade IV) is the most common malignant brain tumor in adults and is associated with a median survival of only 12-15 months despite a multimodal therapy. Glioblastoma is characterized by intratumoral morphological, phenotypic and cellular heterogeneity and among others by the presence of cancer stem cells, which are responsible for tumor formation, recurrence, and resistance to therapy. Cancer stem cells reside in specific perivascular and hypoxic niches. In search for new therapeutic approaches, the focus has recently been shifted to microRNAs, which promote essential pro-tumorigenic properties such as angiogenesis, differentiation and migration and support cellular adaptation to hypoxia.The aim of this work was to identify new key mechanisms that promote tumor malignancy in glioblastomas through hypoxia. Here, we could identify an important pro-tumorigenic role of the hypoxia /HIF induced miR210 in glioblastoma. Methodically, miR210 expression was modulated using lentiviral gain and loss-of function epxeriments in vitro and in vivo in human primary and established glioblastoma cells under normoxic (21% O2) and hypoxic (1% O2) conditions. The over-expression of miR210 promoted stem cell properties in glioblastomas and increased the expression of the tumor stem cell marker CD133 at the protein and /or mRNA level. Concomittantly, miR210 activated the expression of HIF-1a, HIF-2a and specific HIF target genes (VEGF, carbonic anhydrase IX). In line with a pro-tumorigenic role of miR210, the over-expression of miR210 resulted in a significant increase in tumor volume and vascularization in two intracranial xenograft mouse models (GBM046X, G55 GBM). Conversely, the inhibition of miR210 via lentiviral over-expression of antisense oligonucleotides decreased the tumor stem cell phenotype and tumor growth.The key results of this study are, that miR210 plays an important pro-tumorigenic role in glioblastoma and furthermore that the HIF-1 mediated miR210 expression - as part of a feedforward loop- drives HIF dependent tumor characteristics such as angiogenesis or the tumor stem cell phenotype, which are essential for tumor progression. Our findings suggest that miR210 could be used as a potential therapeutic approach for targeted elimination of glioma stem cells in hypoxic niches as well as for the inactivation of the pro-tumorigenic HIF signaling pathway.en
