dc.description.abstractDie Tripeptidylpeptidase 1 (TPP1) ist ein lysosomales Enzym. Es wird als inaktive Vorstufe synthetisiert und durch sauren pH im Lysosom aktiviert. Die Kristallstruktur von TPP1 ist kugelförmig, sein Gen liegt in der Chromosomen-Region 11p15.4. Die seltene lysosomale Speichererkrankung Neuronale Ceroidlipofuszinose 2 (kurz: NCL2, auch CLN2) ist die Folge von Mutationen im TPP1-Gen und wird autosomal-rezessiv vererbt. Die eingeschränkte Funktion des Enzyms TPP1 führt zu Störungen des Substratabbaus und zur intralysosomalen Ansammlung von wachsartigem Ceroidlipofuszin-Material. Dies führt zum weitgehenden Funktionsverlust der Lysosomen. Die Symptome der Klassischen Spätinfantilen Neuronalen Ceroidlipofuszinose zeigen sich erstmals im frühen Kindesalter zwischen 1 und 4 Jahren: psychomotorische Entwicklungsstörungen in Kombination mit epileptischen Anfällen und Demenz durch neurodegenerative Vorgänge. Bereits vorhandene motorische Fähigkeiten gehen verloren und es kommt zum Sehkraftverlust. Meist werden die Patienten nicht älter als 15 Jahre. Bislang ist nur die Enzymersatztherapie für CLN2 zugelassen. Diese ist wenig alltagstauglich, da den Patienten lebenslang etwa alle zwei Wochen rekombinantes TPP1 intravenös, teilweise direkt ins ZNS, verabreicht werden muss. Häufig zeigen sich Immunreaktionen gegen das verabreichte Enzym. Daher wird derzeit nach anderen Therapiemöglichkeiten gesucht, wobei die Faltungstherapie eine Alternative für TPP1-Mutationen darstellen könnte. Die Idee hinter der Faltungstherapie ist, die Strukturveränderungen des mutierten TPP1 zu umgehen, indem die korrekte Faltung des Enzyms unterstützt wird. Hierzu können Chaperone dienen. HSP70 ist ein molekulares Chaperon, das natürlicherweise in den Zellen vorkommt. Es gehört zur sogenannten Hitzeschockantwort, die durch zellulären Stress ausgelöst wird. Als Haupt-Transkriptionsfaktor für Hitzeschockproteine steuert der Hitzeschockfaktor 1 (HSF1) Prozesse im Zellstoffwechsel, die dem zellulären Schutz dienen und reguliert die Transkription der Hitzeschockproteine bei zellulärem Stress hoch. HSP70 agiert als Faltungshelfer und sorgt für die Stabilisierung der Proteine in ihrer Faltungsphase, sodass Fehlfaltungen vorgebeugt werden können. Das Ziel dieser Doktorarbeit war die Untersuchung der Wirkung von HSP70 auf die Expression, Prozessierung und Aktivität von mutiertem TPP1. Es wurden sehr häufig vorkommende TPP1-Missense-Mutationen ausgewählt: Asp276Val, Gly284Val, Ile287Asn, Ser475Leu. Die zugrundeliegenden Zellsysteme waren Patientenfibroblasten und transfizierte HEK293T-Zellen. Um eine Steigerung von HSP70 in den Zellen zu erreichen, wurden die transfizierten Zellen mit den Substanzen Arimoclomol, Celastrol oder mit rekombinantem HSP70 behandelt. Arimoclomol aktiviert die Hitzeschockantwort und sorgt durch die Aktivierung von HSF1 für eine gesteigerte HSP70-Expression. Celastrol ist ein Naturprodukt aus der Wurzel eines ostasiatischen Spindelbaumgewächses. Auch dieses aktiviert HSF1 und bewirkt eine vermehrte HSP70-Präsenz in den Zellen. In dieser Doktorarbeit sollte überprüft werden, ob durch die Behandlung der Zellen mit den genannten Substanzen eine Steigerung von HSP70 und damit verbunden vermehrt korrekt gefaltetes TPP1 erreicht werden kann. Es konnte Folgendes gezeigt werden: Nur bei den Active-Site-Mutanten Asp276Val und Ser475Leu wurde das mutierte TPP1 prozessiert. Bei den anderen Mutanten sammelte sich der TPP1-Vorläufer an. Die Mutation der Active-Site-Mutanten liegt in deren aktivem Zentrum. Der Rest des Proteins wird korrekt aufgebaut und war im Western Blot detektierbar. Da das fehlerhafte aktive Zentrum jedoch keine Substrate binden kann, war auch keine Enzymaktivität vorhanden. Eine Stabilisierung des Faltungsprozesses erzielte bei diesen beiden Mutationen folglich keine Aktivität, da es sich nicht um ein Faltungsproblem handelt. Bei den Mutanten Gly284Val und Ile287Asn hingegen handelt es sich um genau solche Mutationen mit Faltungsfehlern. Diese Mutanten ließen sich unter Umständen durch die Verbesserung des Faltungsprozesses mithilfe von Chaperonen korrigieren. Im Vergleich zur Aktivität des Wildtyps konnte bei diesen beiden Mutanten keine aussagekräftige Aktivität des mutierten TPP1 festgestellt werden (unter 5%). Auch unter Stimulation mit den Substanzen Arimoclomol, Celastrol und rekombinantem HSP70 ergaben sich keine signifikanten Änderungen dieser Ergebnisse. Die Chaperontherapie stellt damit keine Alternative zur zugelassenen Enzymersatztherapie dar. Zumindest für die hier untersuchten Mutationen lieferten die Versuche keine positiven Ergebnisse. Inwieweit die Faltungstherapie auf andere CLN2-Mutationen wirkt, muss noch in weiteren Untersuchungen überprüft werden.de_DE
dc.description.abstractThe enzyme tripeptidylpeptidase 1 (TPP1) is a lysosomal aminopeptidase that cleaves tripeptides from the N-terminus of its substrates. TPP1 is synthesized as an enzymatically inactive precursor, which is further cleaved autocatalytically in the acidic environment of the lysosome. The enzyme is trimmed to the 368 C-terminal amino acids during processing and is thereby activated. The crystal structure of the TPP1-molecule is globular and its gene is located on chromosome 11p15.4. The Neuronal Ceroid Lipofuscinosis 2 (CLN2) is a lysosomal storage disorder. It is also known as classic late infantile neuronal ceroid lipofuscinosis. CLN2 is an autosomal recessive disorder. Compromized activity of TPP1 leads to impairment of the cleavage of its substrates and accumulation of waxy ceroid lipofuscin material in lysosomes. This results in a loss of function of the lysosomes. The symptoms of CLN2 appear in early childhood at the age of 1 to 4 years: psychomotor developmental disorders together with seizures and dementia caused by neurodegenerative decline. Cognitive and motor abilities that have been already achieved are lost. Consequently, vision loss is likely to occur. Mortality is high among patients, and death frequently occurs by 15 years.Enzyme replacement therapy is the only one therapy for CLN2 so far, which is not well compatible with everyday life. In the course of the therapy, recombinant TPP1 needs to be administered by intravenous injection every two weeks, partially directly into the CNS. The drug can possibly trigger immune reactions. For these reasons, we are looking for other treatment options such as folding therapy which might be an alternative for TPP1 mutations that result in misfolding of the protein. The idea of folding therapy is to support the correct folding of the protein and avoid structural changes. Chaperones can be useful, as they are known to support protein folding. HSP70 is a molecular chaperone that occurs naturally in cells. It is part of the heat shock response, which is triggered by cellular stress that can be thermal or chemical, for instance. Heat shock factor 1 (HSF1) is the main transcriptional factor for heat shock proteins. It controls the processes of the cellular metabolism in order to protect the cells. HSF1 up-regulates the transcription of the heat shock proteins like HSP70 in times of cellular stress. HSP70 supports protein folding by stabilizing proteins during this process, which helps to prevent misfolding. The purpose of this work was to investigate the effect of HSP70 on the expression, processing and activity of mutant forms of TPP1. Frequently occurring TPP1-missense-mutations were chosen for the investigation: Asp276Val, Gly284Val, Ile287Asn and Ser475Leu. Experiments were performed in patient fibroblasts and transfected HEK293T cells. To attain more HSP70 in the cells, transfected cells were treated with the substances Arimoclomol, Celastrol or recombinant HSP70. Arimoclomol is an activator of the heat shock response and enhances the HSP70-expression by activation of HSF1. Celastrol is a natural product from the extract of the root of an East Asian spindle tree greenhouse. Celastrol activates HSF1 and thereby a higher HSP70 presence in the cells. These substances are encouraging further studies as therapeutic agents for NCL-diseases. This study aimed at investigating whether treatment of the cells with the previously mentioned substances can trigger an increase of HSP70 and thus enhanced expression of correctly folded TPP1. The following could be shown: Only the active site mutations Asp276Val and Ser475Leu generated processed TPP1. The other mutations showed an accumulation of the precursor. Active site mutations are characterized by an amino acid change located in the active center. Since the remaining part of the protein is built correctly, it can be detected in the Western blot. However, the mutated active site cannot bind substrates. Consequently, there is no remaining enzyme activity. Stabilizing the process of folding did not increase the activity of these two mutations because their functional impairment was not based on a folding error, but direct lack of catalytic activity. However, Gly284Val and Ile287Asn are mutations with a defective folding. If the process of folding could be improved with the help of chaperones, the mutations could theoretically be corrected. However, compared to the wildtype activity, there was no significant activity of the mutant TPP1 to be detected (less than 5%). Treatment with Arimoclomol, Celastrol and recombinant HSP70 did not change these results significantly. In conclusion, chaperone therapy does not represent an alternative to the approved enzyme replacement therapy today. Regarding the mutations tested in this work, the investigations did not provide positive results. Future research should examine to what extent folding therapy is effective for other CLN2 mutations.en
