dc.description.abstractNeovaskuläre Erkrankungen des Auges wie altersbedingte Makuladegeneration (AMD), Frühgeborenenretinopathie (ROP) und diabetische Retinopathie (DR) werden hauptsächlich durch eine hypoxieinduzierte Verstärkung der vascular endothelial growthfactor (VEGF)- Expression hervorgerufen.Die erhöhte VEGF-Konzentration führt zur Bildung von unnatürlich vielen und unreifen Blutgefäßen, die im Auge die visuelle Funktion erheblich beeinträchtigen können. Aktuelle Therapiemaßnahmen zur Regulierung der VEGF induzierten Neovaskularisierung im Auge bedienen sich der monatlichen Injektion von anti-VEGF-Molekülen. Der entscheidende Nachteil dieser anti-VEGF-Therapie besteht jedoch darin, dass ein VEGFAnstieg durch die gängigen Parameter wie Sehschärfe (VA) und die spektrale optische Kohärenz Tomografie (spectral OCT) erst mit einer gewissen Verzögerung erkannt werden kann. Des Weiteren ist eine invasive Probengewinnung notwendig.Das Ziel dieser Studie ist es, ein neues Biolumineszens-Resonanz-Energie-Transfer-System zu etablieren, das sich zur nicht invasiven VEGF Detektion eignet.Genutzt wird die lumineszenten Renilla reniformisform (RLuc8) (lem=~480nm), die in Kombination mit dem Substrat Coelenterazine (CLZ) als Donor dient. Der chemisch generierte Fluorophor, PerCP-Cy5.5® wird als Akzeptor genutzt. Um die Verbindung zu VEGF zu schaffen wird RLuc8 an Ra02, ein Einzelketten variables Fab-Fragment (scFab) eines etablierten anti-VEGF Medikaments, fusioniert, welches an die Rezeptorbindestelle des VEGF bindet. PerCP-Cy5.5® wird an Neuropilin gekoppelt, welches seinerseits an die Neuropilinbindestelle von VEGF bindet.Durch die VEGF-Bindung werden Donor und Akzeptor nun in ausreichende Nähe zueinander gebracht, sodass ein Energieübertrag zwischen ihnen möglich ist, der VEGF nachweist. Anhand der Menge des vom angeregten Fluorophor ausgesendeten Lichts (in BRET-Unit) könnten zusätzlich Rückschlüsse auf die VEGF-Konzentration einer Lösung gezogen werden.Ein Ziel wäre, dass das neue BRET-Paar über ein vivo applizierbares Verfahren immobilisiert in implantierbaren Kapseln die aktuelle VEGF-Konzentration im Auge misst. So wäre es möglich eine, den Patientenbedürfnissen angepasste anti-VEGF-Dosis zu bestimmen. Die Therapie von neovaskulären Erkrankungen könnte folglich deutlich verbessert werden.de_DE
dc.description.abstractNeovascular diseases of the eye, such as age-related macular degeneration (AMD), retinopathy of prematury (ROP) and diabetic retinopathy (DR) are caused by a hypoxia-induced upregulation of vascular endothelial growth factor (VEGF) expression. The resulting excessive supply of VEGF leads to the formation of an unusual and unnatural number of new and immature blood vessels in the eye which may impair visual function.To avoid the increasing development of immature vessels in the eye, the current state-ofthe- art-treatment is a monthly injection of anti-VEGF molecules like Avastin®, Lucentis® and EYLEA®, which prevent VEGF binding to the VEGF-receptors and thus leading to a decreasing activation of VEGF-induced intracellular signaling. However, the crucial disadvantage of the anti-VEGF-therapy lies in the delayed indication of a decreasing VEGF inhibition by diagnostic parameters, such as visual acuity (VA) and ocular spectral optical coherence tomography (spectral OCT), which are indispensable for monitoring the need and success of the therapy. Furthermore, the invasive sampling poses distinct risks to the patients.The aim of this study is to turn the Bioluminescence Resonance Energy Transfer System into standard for non-invasive VEGF detection. Therefore, a luminescent VEGF binding molecule (donor), which binds to the receptor-binding motif of VEGF, was created. This donor transfers energy via BRET to a fluorophore-coupled VEGF binding molecule (acceptor) that binds to the respective neuropilin binding motif of VEGF. The emission of the acceptor appears at a longer wavelength than the donor, thus leading to an increased BRET signal only when VEGF is bound to both, the donor and acceptor molecules. In detail, Renilla reniformis mutant (RLuc8, lem=~480nm) was fused to a single chain variable fragment (scFv) based on Ranibizumab (Lucentis®) named as Ra02, which binds to the receptor binding motif of VEGF. The fluorophore PerCP-Cy5.5® was coupled to neuropilin which binds to the neuropilin motif of VEGF. The implantation of capsules consisting of the two BRET components in solution, permeable for VEGF for its in vivo detection, would provide a new and improved method for monitoring VEGF-induced pathologies and thereby an adjustment of therapy to patient needs.en
