Charakterisierung von Antikörpern gegen P-12-LOX und Aufbau von Nachweisverfahren (ELISA und Durchflusszytometrie)

dc.description.abstract | P-12-LOX gehört zur Gruppe der Dioxygenasen und katalysiert die Reaktion von Arachidonsäure zu 12-HETE. P-12-LOX wird bei verschiedenen Krebserkrankungen vermehrt exprimiert. Es liegen Hinweise vor, dass Produkte von P-12-LOX-katalysierten Reaktionen eine Rolle bei Tumorentwicklung, Tumorwachstum und Metastasierung spielen. Bisher gab es keine Möglichkeit, P-12-LOX mit Hilfe eines routinefähigen Verfahrens, z.B. eines ELISA, nachzuweisen. Grund hierfür war das Fehlen eines geeigneten Antikörperpaares für P-12-LOX.In der vorliegenden Arbeit wurde ein ELISA-Test zum Nachweis von P-12-LOX in Körperflüssigkeit und Gewebeextrakten entwickelt. Der Test wurde auf seine Eignung für den quantitativen Nachweis von P-12-LOX geprüft.Darüber hinaus wurde untersucht, ob P-12-LOX mittels Durchflusszytometrie in Thrombozyten nachgewiesen werden kann.Die Spezifität von anti-P-12-LOX Antikörpern wurde detailliert untersucht. Mit Hilfe zweier ausgesuchter Antikörper (polyklonaler Antikörper IgG 7212 und monoklonaler Antikörper 7213 mAb) wurde ein sog. Sandwich-ELISA aufgebaut. Der ELISA hat eine ausreichende Sensitivität und Genauigkeit für den Einsatz unter Routinebedingungen. Der ELISA wird durch die Probenmatrix Plasma praktisch nicht beeinflusst und kann zur Quantifizierung von P-12-LOX in Plasma eingesetzt werden. Nach Permeabilisierung mittels Detergenzbehandlung ist es mit den charakterisierten Antikörpern möglich, P-12-LOX in Thrombozyten durchflusszytometrisch nachzuweisen.Der entwickelte ELISA-Test ist eine geeignete routinefähige Methode zum Nachweis von P-12-LOX. Auch die Durchflusszytometrie ermöglicht einen Nachweis von P-12-LOX unter Routinebedingungen. Zusammengefasst erlauben die in der vorgelegten Arbeit beschriebenen Verfahren Untersuchungen zur Rolle von P-12-LOX als Biomarker unter physiologischen und pathologischen Bedingungen. | de_DE |
dc.description.abstract | P-12-LOX constitutes a group of dioxygenases. It catalyses the reaction of arachidonic acid yielding 12-HETE. Elevated levels of p-12-LOX are expressed in a variety of tumors. There is evidence that products of the p-12-LOX-catalysed reactions play a role in tumor development, tumor growth and metastatic potential. Up to now the detection of p-12-LOX with routine methods, such as for example ELISA, was not feasible.In this dissertation, an ELISA-test for the detection of p-12-LOX in body fluids and tissue extracts has been developed and checked for its ability to quantitate p-12-LOX. Moreover, it has been explored whether p-12-LOX can be detected in platelets by flow cytometry.Reactivity and specificity of several anti p-12-LOX antibodies was explored in detail. By using two selected antibodies (polyclonal antibody 7212 and monoclonal antibody 7213) a sandwich-ELISA-test was established. Sensitivity and precision of the ELISA were found to be sufficient for routine conditions. Moreover, basic reactivity of the ELISA-test was not found to be affected by the sample matrix plasma. In conclusion the ELISA can be used for quantification of p-12-LOX in blood plasma.Anti p-12-LOX antibodies were also used for flow cytometry. A staining procedure with a permeabilization step was developed that allows detecting intracellular p-12-LOX in platelets by flow cytometry.Taken together, the methods developed in this dissertation are important tools for research on the role of p-12-LOX as a biomarker under physiological und pathological conditions. | en |
