dc.description.abstractOsteoporose gilt als einer der Hauptrisikofaktoren für eine verzögerte Frakturheilung, weshalb neue Knochenersatzmaterialien auf eine Verbesserung dieser zielen. CPC ist durch seine Osteokonduktivität und Biokompatibilität als Grundbaustein geeignet, und wurde in dieser Studie zur Stimulation der Knochenneubildung im murinen metaphysären Frakturspalt in Form von pastösem alpha-TCP-basiertem HA-bildendem Knochenzement mit BDNF-gekoppelten MBG-Partikeln versetzt. ToF-SIMS, REM / EDX und Lichtmikroskopie wurden zur Auswertung der Knochen-Zement-Grenzfläche benutzt.ToF-SIMS-Messungen konnten verschiedene Material- und Gewebeverteilungen in räumlichem Bezug zueinander darstellen. Im Vergleich mit EDX-Mappings und lichtmikroskopischen Aufnahmen zeigten diese, dass an der Grenzfläche und in Poren des mit BDNF funktionalisierten CPC-MBG-Komposits sowohl bei WT- als auch bei osteoporotischen M3R-KO-Mäusen eine Knochenneubildung bestehend aus unmineralisiertem und bereits mineralisiertem Gewebe stattfand. Durch Auflösung von MBG-Partikeln und daraus resultierender Porenbildung wurde der Zementabbau und die knöcherne Integration an entsprechenden Stellen beschleunigt. Das meiste Material war nach 35 Tagen zum Ende des Experiments jedoch noch vorhanden. Mithilfe der chemischen Verteilung der ToF-SIMS und EDX-Untersuchungen konnte nachgewiesen werden, dass peripher im Zement liegende MBG-Partikel sich durch Kontakt mit dem Knochenmark auflösen und resorbiert werden konnten. Durch den hier verwendeten Anteil an MBG-Partikeln von 2 wt% lagen vor allem in der osteoporotischen Gruppe jedoch fast keine randständigen Partikel vor, weshalb die BDNF-Freisetzung aus dem Material als nicht ausreichend einzustufen ist. Aus diesem Grund konnten auch durch das Ca / P-Verhältnis bei der EDX-Quantifizierung und durch die Osteocytenauswertung keine deutlichen Unterschiede in den Gruppen mit BDNF im Vergleich zur Kontrollgruppe ohne BDNF erkannt werden. An Poren, wo von einer BDNF-Wirkung ausgegangen werden kann, waren keine auffälligen Veränderungen der Osteocyten sowie der Mineralisierung sichtbar.Zum einen kann schlussgefolgert werden, dass ein zu geringer Anteil an MBG (+ BDNF) dem CPC beigemischt wurde, zum anderen ist dadurch keine sichere Aussage über einen positiven Effekt von dem daher ebenfalls zu wenig freigesetzten BDNF möglich. Um eine deutliche Wirkung auf die Frakturheilung erzielen und diese beurteilen zu können, sollten deshalb in zukünftigen Studien deutlich mehr Partikel mit höheren Konzentrationen an BDNF als in dieser Studie möglich waren, dem Zement beigemischt werden.de_DE
dc.description.abstractOsteoporosis is considered to be one of the major risk factors for delayed fracture heal-ing, therefore new implant materials are targeting the osteoinductive effect. Because of its osteoconductivity and biocompatibility CPC is a suitable material, which contains BDNF-coupled MBG particles to stimulate new bone formation in the murine metaphyseal fracture gap in the form of a pasty alpha-TCP-based HA-forming bone cement in this study. ToF-SIMS, REM / EDX and histological stainings were used to evaluate the interface of bone and implant material.ToF-SIMS measurements were able to show distributions of different materials and tissues in spatial relation to each other. Compared with EDX mappings and histological analyses ToF-SIMS measurements revealed that bone formation of unmineralized bone matrix and partly mineralized tissue occurred at the interface and infiltrated the pores of the BDNF-functionalized CPC-MBG composite in both WT and osteoporotic M3R-KO mice. The dissolution of MBG particles and resulting pore formation accelerated cement removal and bone integration. However, most of the material was still present at the end of the study after 35 days. The chemical distribution of the ToF-SIMS and EDX measurements revealed that MBG particles located peripherally in the cement could dissolve and be resorbed by contact with the bone marrow. Due to the amount of MBG particles of 2 wt% which was used here, especially in the osteoporotic group almost no marginal particles were detected, so the BDNF release can be classified not to be sufficient. For this reason, also the Ca / P ratio of EDX quantification and the evaluation of osteocytes did not show any clear differences in the groups with BDNF compared to the control groups without BDNF. In pores where a release of BDNF can be expected no obvious changes in osteocytes and mineralization were seen.Therefore, it can be concluded that, on the one hand the amount of MBG (+ BDNF), which was added to the CPC, was not enough and, on the other hand, no reliable state-ment about a positive effect of BDNF, which is also under-released, is possible. In order to achieve a clear effect on the fracture healing, in future studies more particles with higher concentrations of BDNF that was possible in this study should be added to the cement.en
